小麦及其近缘种中Yr10第一外显子功能标记初步研究

摘  要:条锈病是小麦生产上危害最严重的病害之一。抗条锈病基因Yr10是抗性优良且在生产上还未被大量利用的基因。为给Yr10基因的筛选及利用提供理论参考和技术支撑,以对条锈菌抗性不同的小麦及其近缘种共28个材料,采用PCR扩增技术对Yr10基因的第一外显子进行了功能标记初步研究。结果表明:小麦抗性材料中不含有Yr10基因,这些抗性材料中的抗性由其他抗性基因决定;感病材料26号(提莫菲维)的基因组中含有与Yr10基因第一外显子序列相似性较高的等位基因或抗性相关等位基因,该材料中扩增产物与Yr10中在决定氨基酸的关键位点上应该存在序列差异,进而导致抗性上的对立。Stripe rust (Puccinia striiformis f sp. tritici ) is the most serious diseases of wheat production.Wheat stripe rust resistance gene YrlO is a good resistance gene that had not been widespread applied.The study selected wheat and related species total of 28 materials of different resistance, used PCR to amplify the first extron, for gene Yr10 screening and utilizingto provided theoretical reference and technical support.The result showed that there wasn't Yr10 gene in wheat materials in the materials in the study. The resistence of wheat and related species materials of the study might be decided by other resistance genes.In the genome of the susceptible 26(Timo Feiwei) contains the very similar sequence with the first exon of gene Yr10 alleles or resistance-related alleles,the material in the amplified product eompared with the Yr10 critical sites of determining the amino acid sequence differences should exist differences, which led to the opposition of the resistance.
