猪ATIC基因SNPs位点分析和表达规律的研究

摘　　要：为了进一步探索氨基咪唑氨甲酰核苷酸转甲酰基酶/次黄嘌呤核苷酸环水解酶ATIC基因与肉质风味性状的关系,寻找猪肉质风味性状相关分子标记,研究采用PCR-SSCP的方法对ATIC基因进行序列扫描,寻找SNP位点并分析不同等位基因在不同品种猪间的分布规律,同时采用荧光定量PCR的方法对ATIC基因在猪子宫、背肌、肠、肺、脾脏、肝脏、心脏、脂肪、腿肌等共9种组织器官中的表达情况进行研究。结果表明,各等位基因在民猪、北京黑猪、长白猪、大白猪等猪种间的分布存在着极显著差异(P<0.01)。ATIC基因在肝脏、腿肌、肺中高表达,其中在肝脏中的表达量最高。In order to further the relationship between 5-aminoimidazole-4-carboxamide ribonucleotide formyltransferase/IMP cyclohydroase（ATIC） and meat quality of flavor characteristic trait,find the molecular marker of pork quality flavor characteristic traits.In the paper,the method of PCR-SSCP was used to look for SNPs in ATIC and analyze the distribution regularity of different alleles in various species of pigs.At the same time,analysis on the expression of ATIC gene in 9 tissues including uterine,back muscle,bowel,lung,spleen,liver,heart,fat and leg muscles by RT-PCR.The distribution of various alleles exist extreme difference（P0.01） in Min Pig,Beijing Black Pig,Landrace,Yorkshire.ATIC was highly expressed in liver,led muscles and lung among which expression had the highest value in lung.
