摘  要:目的比较复合不同剂量血管内皮生长因子(VEGF)的纳米羟基磷灰石/胶原蛋白复合骨(NHAC)修复骨缺损的效果。方法将纳米羟基磷灰石粉末和胶原蛋白粉末按8∶2的比例混合制备NHAC人工骨,再将混合粉末与含10 ng、100 ng、300 ngVEGF的蒸馏水按1∶1.8的质量比调和制备NHAC/VEGF人工骨。建立大鼠双侧桡骨0.5 cm缺损动物模型30只,按随机原则分为5组。以NHAC/10 ngVEGF、NHAC/100 ngVEGF、NHAC/300 ngVEGF人工骨植入骨缺损处进行修复作为实验组,以NHAC人工骨植入组及空白组作为对照组。术后2、4、8周各组行组织学及免疫组织化学检查,观察材料早期血管化及成骨情况。结果各时间点组织学评分以及血管计数均为NHAC/300 ngVEGF人工骨组最高,于其他各组比较差异有统计学意义。各实验组血管数量在4周时达到高峰,而NHAC组血管数量在8周时最多。结论血管内皮生长因子能明显促进NHAC早期血管化及新骨形成,比单纯应用NHAC能更好的修复骨缺损,并且随着VEGF剂量的增加,新生血管数量和新骨形成量均相应增加。Objective To investigate the effect of nano-hydroxyapatite/collagen (NHAC) materials combined with variant doses of vascular endothelial growth factor ( VEGF ) for the repair of bone defects. Methods Nano-hydroxyapatite powder and collagen powder were mixed in the ratio of 8:2 and prepared into NHAC artificial bone. The mixed powder was mixed with 10 ng, 100 ng and 300 ng of VEGF respectively in the ratio of 1:1.8, and were then prepared into NHAC/VEGF artificial bone. Thirty healthy rats whose radiuses were cut off 0.5 cm were divided into five groups (twelve radiuses in each group). Groups in which bone defects of rats were repaired with NHAC/10 ngVEGF,NHAC/100 ngVEGF and NHAC/300 ngVEGF artificial bones were the experimental groups. The NHAC group and the blank group were the control groups. The vasculm'ization of artificial bone implanted into the defect area and new-bone formation were evaluated by histological and immunohistochemical analysis at 2,4 and 8 weeks postoperatively. Results The new bone formation,molding histological score and blood vessel count at each time point of NHAC/300 ngVEGF group were significantly higher compared to those of the other groups ( P 〈 0.05 ). Blood vessel counts in the experimental groups reached the maximum at 4 week, while the NHAC group at 8 week. Conclusion VEGF is beneficial tot promoting early vascularization of the NHAC and improves the repair of bone defects, and the blood vessel count and new bone tonnation increase as VEGF increases.
