摘 要:本文采用鹿角菜胶诱导的大鼠急性炎症模型观察橄榄叶提取物(OLE)的抗炎与镇痛作用及对致炎因子TNF-α和IL-1β的mRNA表达。用鹿角菜胶注射到大鼠右后足内,分别于鹿角菜胶注射前30 min和注射后120 min灌胃给予OLE 100、250和500 mg/kg体重。鹿角菜胶处理后测量足跖肿胀度及痛域。用RT-PCR检测足跖垫内组织TNF-α、IL-1β和IL-10的mRNA表达。结果显示,经灌胃给予OLE100、250和500 mg/kg体重,均能明显降低大鼠后足肿胀度,增加痛域,并明显降低致炎因子TNF-α和IL-1β的mRNA表达。本研究提示OLE具有抗炎和镇痛作用,其作用机制可能与抑制TNF-α和IL-1β的mRNA表达有关。The aim of this study was to assess the inhibitory role of olive leaves extract (OLE) on carrageenan-induced swelling and hyperalgesia of rat paw. Paw inflammation was assessed by the increase in paw volume and hyperalgesia. The levels of the tumor necrosis factor alpha (TNF-α) ,interleukin 1/3 (IL-1β) and interleukin 10 (IL-10) mRNA in the tissue of right paw were estimated by reverse transcription-polymerase chain reaction (RT-PCR). The results showed that the paw pressure thresholds of rats orally administered OLE significantly increased the degree swelling of right hind paw showed a statistically significant reduction, compared with rats in model control. OLE decreased expression of pro-inflammatory cytokines IL-1β and TNF-α. mRNA The results of the study suggest that OLE could have anti-inflammatory and anti-hyperalgesic effects and the mechanisms of these effects may be involved in inhibition of TNF-α and IL-1β mRNA expression.
