摘  要:目的探讨丝裂原活化蛋白激酶(MAPK)中的细胞外调节蛋白激酶(ERK)在低氧性肺动脉高压中的表达以及三七总皂苷(PNS)对慢性低氧性肺动脉高压(HPH)大鼠ERK表达的影响。方法将30只SD大鼠随机分为3组:正常对照组(N组)、单纯低氧性肺动脉高压组(H组)和低氧性肺动脉高压注射PNS组(HP组)。观察各组大鼠平均肺动脉压(mPAP)及平均颈动脉压(mCAP),免疫组化法检测肺组织中p-ERK蛋白的含量。结果 H组、HP组ERK的表达高于N组(P<0.05),且H组高于HP组(P<0.05)。结论预防性应用PNS可以降低低氧性肺动脉高压大鼠的肺动脉压,其机制可能与抑制p-ERK的表达有关。Objective To investigate the expression of ERK in MAPK in chronic hypoxic pulmonary hypertension,and the effect of the panax notoginseng saponins (PNS) on the expression of ERK. Methods 30 SD rats were randomly divided into 3 groups :normal con- trol group (N group) , simple hypoxic pulmonary hypertension group (H group) and hypoxic pulmonary hypertension injected PNS group (PNS group). The mean pulmonary artery pressure (mPAP) and mean carotid arterial pressure (mCAP) of rats in each group were ob- served,and immunohistochemistry method was used to detect the ERK protein content in lung tissues. Results The expression of ERK in H Group and HP Group was higher than that of N group (P 〈 0.05). Moreover the H group was higher than the HP group (P 〈 0.05 ). Conclusion The prophylactic application of PNS can reduce the pulmonary artery pressure of the hypoxic pulmonary hypertension in rats, and its mechanism may be related to the inhibition of p - ERK expression.
