空肠弯曲菌pcDNA3.1(-)-peb1A诱导的T细胞活化增殖研究

摘　　要：目的探讨空肠弯曲菌(Campylobacter jejuni,CJ)pcDNA3.1(-)-peb1A疫苗免疫昆明种小鼠后T细胞活化增殖状态。方法重组质粒CJ pcDNA3.1(-)-peb1A免疫昆明种小鼠后,取其脾淋巴细胞,应用ELISA法检测其膜表面分子CD3、CD4、CD8、CD25和CD28。结果检测结果显示:CD3检测,佐剂DNA组高于裸DNA组,在第10天有显著性差异(P<0.05);CD8检测,佐剂DNA组高于裸DNA组,在第10、20天均有显著性差异(P<0.05),裸DNA组也高于两对照组,但仅在第20天有显著性差异(P<0.05);CD25检测,裸DNA组高于NS组,但仅在第10天有显著性差异(P<0.05),佐剂DNA组高于裸DNA组,也仅在第20天有显著性差异(P<0.05);CD4及CD28检测,佐剂DNA组高于裸DNA组,在第10、20天均有显著性差异(P<0.05);在第10、20天各脾淋巴细胞膜表面分子检测,佐剂DNA组高于两对照组,有显著性差异(P<0.05)。结论 CJ pcDNA3.1(-)-peb1A疫苗能够有效地诱导T细胞活化增殖。We aimed to explore activation and proliferation of T lymphocyte induced by the DNA vaccine pcDNA3.1（-）-peb1A against CJ on the Kunming mouse.Lymphocytes was gathered from spleens of the Kunming mice immunized with pcDNA3.1（-）-peb1A and then we assayed their surface molecules CD3,CD4,CD8,CD25 and CD28 with the method of enzyme-linked immunosorbent assay（ELISA）.The results showed that：on the assay of CD3,the adjuvant DNA group was higher than the bare DNA group and it had a significant difference on the 10th day（P0.05）;On the assay of CD8,the adjuvant DNA group was higher than the bare DNA group and it had a significant difference on the 10th and 20th day（P0.05）,the bare DNA group was higher than the two control groups but it had a significant difference only on the 20th day（P0.05）;On the assay of CD25,the bare DNA group was higher than the NS group but it had a significant difference only on the 10th day（P0.05）,the adjuvant DNA group was higher than the bare DNA group but it had a significant difference only on the 20th day（P0.05）;the adjuvant DNA group was higher than the bare DNA group and it had a significant difference on the 10th and 20th day of CD4 and CD28（P0.05）;The adjuvant DNA group was higher than two control groups and it had a significant difference on the 10th and 20th day of all the lymphocytes surface molecules（P0.05）.The results showed that CJ pcDNA3.1（-）-peb1A vaccine could induce the effective activation and proliferation of T lymphocyte.
