摘 要:采用10种含有不同植物激素的愈伤组织诱导培养基,研究了4种骨干玉米自交系的愈伤组织诱导情况。结果表明,基因型对愈伤组织诱导率影响显著,且基因型和培养基之间存在互作;2,4-D对玉米幼胚愈伤组织的诱导是必需的,2.0~3.0mg/L的2,4-D大多能诱导出胚性愈伤组织;幼胚长为1.5~2.0mm时,玉米愈伤组织诱导率较高且质量较好;10mg/L的AgNO3可以明显提高愈伤诱导率,6-BA对愈伤组织的诱导率没有明显影响。Ten culture media with different plant hormones for callus induction were designed, in order to analyze the callus induction conditions of four excellent maize inbred lines. The results showed that genotypes had significant effects on callus induction, and there were interaction between genotype and medium;2,4-dichlorophenoxyacetic acid(2,4-D) was necessary for callus in- duction,and the optimal concentration for embryogenic callus formation was 2.0--3.0 mg/L;The length of embryo between 1.5--2.0mm was the best time, when higher induction frequency and better quality of embryo callus were gotten;The addition of 10 mg/L AgNO3 to the N6 medium could obviously stimulated embryo callus formation, and the addition of 6-BA had no significant effect on induction frequency.
