摘  要:目的:观察远端缺失同源盒5(distal-less homeobox5,DLX5)、核心结合蛋白因子2(runt-related transcription factor-2,RUNX2)、成骨细胞特异性转录因子Osterix(OSX)和牙本质涎蛋白(dentin sialoprotein,DSP)在出生后SD大鼠下颌第一磨牙不同发育阶段的表达情况,探讨其在SD大鼠出生后下颌第一磨牙发育矿化的作用机制。方法:采用免疫组化法分析出生后1、7、14、28d SD大鼠下颌第一磨牙中DLX5、RUNX2、OSX、DSP的表达分布。结果:DLX5在出生后1、7、14d中均有表达,表达量呈现出上升趋势,但是在28d时不表达;RUNX2在出生后1、7、14、28d均表达,表达量在7d时呈现最低;OSX在出生后1、7、14、28d均有表达,表达量在14d时呈现最低;DSP在出生后1、7、14、28d均有表达,表达量呈上升趋势。结论:DLX5、RUNX2、OSX、DSP在大鼠出生后各阶段的表达量不同,具有时空特异性,表明DLX5、RUNX2、OSX、DSP可能对出生后SD大鼠下颌第一磨牙发育及矿化具有一定的调控作用。Objective: To observe the expression of distall-less homeobox 5 ( DLX5 ), runt-related transcription factor 2 ( RUNX2 ), Osterix (OSX) and dentin sialoprotein (DSP) in mandibular first molars of postnatal (PN) SD rat. Methods: SD rat mandibles were isolated and fixed in 4% polyoxymethylene at PN 1, 7, 14, 28 days respectively. Tissue sections from paraffin blocks were processed for HE and immunohistochemistry staining to detect the expression of DLXS, RUNX2, OSX and DSP. Results :The expression of DLX5 was detected in specimen at PN 1,7, 14 days and displayed an uprising positive staining, but negative at PN 28 days. RUNX2 was de- tected in specimen at PN 1, 7, 14 and 28 days, among which the expression level was the lowest at PN 7 days. OSX was detected in specimen at PN 1, 7, 14 and 28 days, among which the expression level was the lowest at PN 14 days. An uprising trend of OSX levels was detected in specimen at PN 1,7, 14 and 28 days. Conclusions : The expression of DLXS, RUNX2, OSX and DSP in postnatal SD rat mandibular first molar was time-specific, reflecting their different functional state following the odontogenesis.
