摘 要:目的:探讨肝细胞生长因子(hepatocyte growth factor,HGF)对体外培养的根尖牙乳头干细胞(stem cells from apical pa-pilla,SCAP)增殖和矿化能力的影响。方法:将SCAP细胞分别在标准培养液、含有10ng/ml HGF的培养液以及矿化诱导培养液中培养,分为空白对照组、HGF组、矿化诱导组和HGF+矿化诱导组。通过MTT法、碱性磷酸酶(alkaline phosphotase,ALP)活性检测、茜素红染色和氯代十六烷基吡啶定量检测SCAP细胞增殖和矿化能力的改变。结果:MTT结果显示,HGF可以促进矿化诱导下的SCAP细胞的增殖能力(P<0.005)。与其他组相比较,HGF+矿化诱导组SCAP细胞的ALP活性明显上调,矿化能力显著提高(P<0.005)。结论:10ng/ml HGF可明显促进SCAP细胞的增殖和矿化能力。Objective:To observe the effects of hepatocyte growth factor (HGF) on the proliferation and mineralization polential of human stem cells from apical papilla (SCAP) in vitro, nethods:SCAP were treated with 10 ng/ml HGF in normal medium and miner- alization-inducing medium for certain time periods. Proliferation of SCAP was evaluated by MTT colorimetric assay. Alkaline phosphotase (ALP) activity was determined by enzyme kinetics methods. Alizarin red staining and cetylpyridinium chloride (CPC) assay were performed to investigate the mineralization ability of SCAP. Results:The proliferation activity of HGF-treated SCAP in mineralization- inducing medium was significantly higher than other groups ( P 〈 0. 005 ). ALP activity and mineralization ability of this group were sig- nificantly upregulated (P 〈 0. 005). Conclusions: 10 ng/ml HGF significantly promotes the proliferation and mineralization of SCAP, especially in the mineralization-inducing medium.
