摘 要:研究旨在分析乐至黑山羊与藏山羊的热休克蛋白70.1(Hsp70.1)基因之间的差异.利用PCR对乐至黑山羊、藏山羊的Hsp70.1基因进行克隆并测序,并将该基因的氨基酸序列与其他属种进行聚类分析.结果表明,扩增出的乐至黑山羊、藏山羊Hsp70.1基因编码序列均长1926bp,均编码642个氨基酸,两者之间有99.79%的相似度.通过系统发育分析,乐至黑山羊、藏山羊和山羊为一支,肩峰牛和欧洲牛为一支,人单独为一支,褐家鼠单独为一支,非洲爪蟾单独为一支,原鸡单独为一支.The objective of this study is to identify the differences of Hsp70.1 gene between Lezhi black goat and Tibetan goat.The Hsp70.1 gene of Lezhi black goat and Tibetan goat is cloned and sequenced by using the PCR procedure.The deduced amino-acid sequences are compared with other Genus using cluster analysis software.The result shows that the amplification coding region of the Hsp70.1 gene in Lezhi black goat and Tibetan goat is 1926bp long,encoding 642 amino acids.Nucleotide sequence similarities of the Hsp70.1 gene in Lezhi black goat and Tibetan goat is 99.79%.Phylograms constructed on the basis of Hsp70.1 mature peptide sequences from various species show that Lezhi black goat,Tibetan goat and Capra hircus comprise one clade,Bos indicus and Bos taurus comprise another.Homo sapiens,Rattus norvegicus,Xenopus laevis and Gallus gallus represent entirely different clades.
