平喘方及其拆方对支气管哮喘小鼠肺组织转录因子T-bet/GATA-3表达的影响

摘　　要：目的:观察平喘方及其拆方对支气管哮喘模型小鼠转录因子T-bet、GATA-3表达的影响,研究平喘方及其拆方防治支气管哮喘的作用机理。方法:通过对BALB/c小鼠用卵蛋白等腹腔注射致敏、雾化吸入激发,建立支气管哮喘动物模型。将120只清洁级雄性BALB/c小鼠,随机分为6组:对照组、模型组、平喘方组、拆方1组、拆方2组、拆方3组。各组分别于雾化激发哮喘后7d、21d分两批处死、取样、检测,动态观察。采用RT-PCR(RealTimePCR)法检测小鼠肺组织T-bet、GATA-3mRNA的表达。结果:哮喘激发7d,拆方1、2、3组T-bet升高(P<0.05);平喘方组和拆方2、3组GATA-3降低(P<0.05);拆方3组T-bet/GATA-3比值升高(P<0.05)。哮喘激发21d,平喘方及拆方1、2、3组T-bet升高(P<0.01),GATA-3降低(P<0.01),T-bet/GATA-3比值升高(P<0.01);其中平喘方组、拆方3组T-bet表达高于拆方1、2组(P<0.05),GATA-3低于拆方1、2组(P<0.01),T-bet/GATA-3的比值高于拆方1、2组(P<0.05或P<0.01)。结论:平喘方及拆方各组均能不同程度地调节支气管哮喘小鼠Th1/Th2上游因子T-bet/GATA-3mRNA的表达,对Th1/Th2细胞的失衡有纠正作用,拆方3组疗效接近平喘方组。Objective：To discuss effects and mechanism of Ping Chuan formula and its different disassembled formulae on preventing and curing asthma. Methods：To establish the model of asthmatic mice, the BALB/c mice were sensitized by intra-peritioneal injection of mixed ovalbumin and then activated by inhaling ovalbumin. 120 BALB/c mice were randomly divided into 6 groups： the normal group, the asthma model group, the Ping Chuan formula group, CF 1 （Ping Chuan formula disassembled prescription 1 group）, CF2, CF3. After the 7 days and 21 days of activated asthma, the lung cells were treated with Trizol for extraction of total RNA, the gene expression of T - bet, GATA - 3 were analyzed by RT - PCR ; the general histological changes of lung tissue were observed by hematoxylin and eosin stain. Results：At the 7th day, the expression of T - bet in CF1, CF2 and CF3 were obviously increased （P 〈 0. 05 ） ;the expression of GATA - 3 in Ping Chuan formula, CF2 and CF3 were obviously decreasedP 〈 0. 01 ）. The ratio of T - bet/GATA - 3 in CF3 were higher than the other goupe （ P 〈 0. 05 ）. At the 21 th day, the expression of T - bet were increased in every groupe （ P 〈 0. 01 ）, the expression of GATA -3 significantly decreased（P 〈 O. 01 ） ,The ratio of T - bet/GATA - 3 were increased （ P 〈 0. 01 ）. CF1 and CF2＇ s T - bet expression level were lower than Ping Chuan formula and CF3 （ P 〈 0. 05 ）, GATA - 3 were higher （ P 〈 0. 01 ）, and the ratio of T - bet/ GATA -3 were lower than Ping Chuan formula and CF3 （P 〈 0. 05 orP 〈 0. 01 ）. Conclusion：Ping Chuan formula and its different disassembled prescription can regulate the expression of T - bet/GATA - 3 mRNA which are the upstream factors of Th1/Th2, then regulate the balance of Thl/Th2： CF3＇ s curative effect is nearly equal to the effect of Ping Chuan formula.
