摘 要:目的:观察肺岩宁颗粒对Lewis肺癌小鼠细胞周期G1/S检测点信号通路中MDM2-P53生物轴的影响。方法:采用C57BL/6小鼠,造模并随机分为6组:模型组、顺铂组、肺岩宁煎剂组、肺岩宁颗粒低剂量组、肺岩宁颗粒中剂量组、肺岩宁颗粒高剂量组,进行相应药物干预。观察各组抑瘤率、瘤重、体质量、去瘤体质量,流式细胞术检测细胞周期时相分布,RT-PCR测定肿瘤组织MDM2、P53的mRNA表达。结果:肺岩宁颗粒中剂量组瘤重显著低于模型组,差异有统计学意义(P<0.01),与顺铂组比较差异无统计学意义。肺岩宁颗粒中剂量组体质量与模型组比较,差异有统计学意义(P<0.05);与顺铂组比较,体质量明显升高,差异有统计学意义(P<0.01);肺岩宁颗粒中剂量组去瘤后体质量显著高于煎剂组、模型组、顺铂组,差异有统计学意义(P<0.05,P<0.01)。流式细胞术发现肺岩宁颗粒中剂量组的G0/G1比例较模型组和顺铂组显著增多,癌细胞增殖指数也明显降低,差异有统计学意义(P<0.01)。RT-PCR显示肺岩宁颗粒中剂组MDM2 mRNA水平明显低于模型组、顺铂组、煎剂组,差异有统计学意义(P<0.01);肺岩宁颗粒中剂组P53 mRNA表达明显高于模型组、顺铂组和煎剂组,差异有统计学意义(P<0.01)。肺岩宁颗粒高剂量组无小鼠存活,考虑与颗粒剂溶解度有关。结论:在Lewis肺癌模型中,调节细胞周期G1/S检测点信号通路MDM2-P53生物轴可能是肺岩宁颗粒抗肺癌增殖的重要途径之一。Objective:To observe the effects of "Feiyanning Granule" on MDM2-P53 bio-axis of Lewis lung cancer cell cycle G1/S checkpoint signal pathway.Methods:C57BL/6 mice were applied,and the Lewis lung cancer model mice were established.The model mice were randomly divided into six groups:model group(A),cisplatin group(B),"Feiyanning" decoction group(C),"Feiyanning Granule" low-,medium-and high-dose groups(D,E,F),with corresponding drug administered.The tumour inhibiting rate,tumour weight,body weight and body weight without tumour were observed.Cell cycle distribution was detected by flow cytometry(FCM),and the expressions of MDM2 and P53 mRNA in tumour tissues were tested by RT-PCR.Results:The tumour weight in group E was lower than that in group A(P〈0.01),and had no statistical difference compared with group B.The body weight of group E was significantly higher than that of groups A and B(P〈0.05,P〈0.01);the body weight without tumour of group E was obviously higher than that of groups A,B and C(P〈0.01,P〈0.05).The FCM analysis showed that the proportion of G0/G1 phase cell cycle in group E was increased,and the cancer cell proliferation index(PI) was decreased compared with groups A and B(P〈0.01).The RT-PCR results showed that the level of MDM2 mRNA expression of group E was lower than that of groups A,B and C(P〈0.01),and the level of P53 mRNA expression of group E was higher than that of groups A,B and C(P〈0.01).There were no mice alive in group F,and the death had relationship with granule solubility.Conclusion:Regulating the MDM2-P53 bio-axis of cell cycle G1/S checkpoint signal pathway is one of the important pathways for "Feiyanning Granule" to inhibit lung cancer cell proliferation in Lewis lung cancer mice.
