摘 要:目的观察地灵丹对大鼠高尿酸血症肾病的影响,并探讨相关作用机制。方法采用高嘌呤饲料制作高尿酸血症肾病大鼠模型,分为地灵丹治疗组、别嘌呤醇组、模型对照组及正常对照组,每组15只大鼠,分别给予相应的干预措施,持续4周。检测大鼠血尿酸、肌酐变化,观察肾脏组织学变化及细胞因子TGF-β1、TNF-α的表达。结果实验第14天,与正常对照组比较,各造模组血尿酸、血肌酐水平显著升高,差异有统计学意义(P<0.05,P<0.01);与模型组比较,地灵丹组及别嘌呤醇组血尿酸、血肌酐水平均显著降低,差异有统计学意义(P<0.01)。实验第28天,与正常对照组比较,模型组及地灵丹组血尿酸、血肌酐水平显著升高,差异有统计学意义(P<0.01),而别嘌呤醇组血尿酸水平出现显著降低,差异有统计学意义(P<0.01);与模型组比较,地灵丹组及别嘌呤醇组血尿酸、血肌酐水平显著降低,差异有统计学意义(P<0.01);与地灵丹组比较,别嘌呤醇组的血尿酸水平显著降低,血肌酐水平显著升高,差异有统计学意义(P<0.01);与正常对照组比较,其余各组TGF-β1、TNF-α阳性面积均显著增多,差异有统计学意义(P<0.01);与模型组比较,地灵丹组与别嘌呤醇组的TGF-β1阳性面积显著减少,差异有统计学意义(P<0.05,P<0.01)。与模型组比较,地灵丹组与别嘌呤醇组大鼠肾脏病理改变减轻。结论地灵丹可减缓高尿酸血症肾病大鼠肾脏损害的作用,其作用机制可能与调节TGF-β1、TNF-α等细胞因子有关。Objective To observe the effects of "Di Ling Pill"(DLD) on hyperuricemic nephropathy in rats and evaluate its possible mechanisms.Methods We carried out this experiment in rats fed with high purine animal food.First,the rats were randomized into DLD group,allopurinol group,model group and normal group,15 rats in each group.Then,every group was intervened with corresponding drugs for 4 weeks.And then,we detected the change of serum creatinine(Scr),uricacid(UA) and renal pathology,and observed the expression of cytokines on the kidney including transforming growth factor-β1(TGF-β1) and tumor necrosis factor-α(TNF-α).Results On the 14th day,compared with normal group,the serum Scr and UA in the other groups increased significantly(P0.05,P0.01);compared with model group,the serum Scr and UA in DLS group and allopurinol group decreased significantly(P0.01).On the 28th day,compared with normal group,the serum Scr and UA in model group and DLD group increased significantly(P0.01),but the UA in the allopurinol group decreased significantly(P0.01);compared with model group,the serum Scr and UA in Dilingdan group and allopurinol group decreased significantly(P0.01);compared with Dilingdan group,the serum UA in allopurinol group decreased but the serum Scr increased(P0.01);compared with normal group,the expression of TGF-β1and TNF-α in the other groups significantly increased(P0.01);compared with model group,the expression of TGF-β1 in Dilingdan group and allopurinol group decreased significantly(P0.05,P0.01).Compared with model group,the pathological changes in kidneys of the rats in the DLD group and allopurinol group mitigated.Conclusion "Di Ling Pill" can protect the renal function of the rats of hyperuricemic nephropathy from injury by modulating cytokine like such as TGF-β1 and TNF-α.
