摘 要:建立了高效液相色谱-电喷雾质谱(HPLC-ESI-MS)法同时测定人体尿液中黄蝶呤与异黄蝶呤的分析方法。尿样经离心、过膜后直接进样,采用电喷雾质谱进行定性和定量。结果表明,黄蝶呤与异黄蝶呤含量分别在0.0204~2.04μg/mL和0.0202~2.02μg/mL范围内与色谱峰面积呈良好的线性关系,线性相关系数r2分别为0.9995、0.9997,检出限分别为0.007μg/mL和0.006μg/mL。黄蝶呤与异黄蝶呤平均回收率在88.0%~110.0%之间。该方法前处理简单,易于操作,为进一步应用到临床癌症早期诊断作了方法学的初步探讨。A novel method was developed and validated for the simultaneous determination of xanthopterin and isoxanthopterin in human urine using liquid chromatography-electrospray ionization mass spectrometry.Urine samples were centrifuged and filtered,then analyzed by electrospray ionization mass spectrometry.The linear ranges of xanthopterin and isoxanthopterin were 0.0204~2.04 μg/mL and 0.0202~2.02 μg/mL,respectively.The linear regression equations were y=342622x-7848(r2=0.9995) and y=309694x-3573(r2=0.9997),respectively.The detection limits were 0.007 μg/mL and 0.006 μg/mL,respectively.For xanthopterin and isoxanthopterin in human urine,the recoveries were in the range of 88.0%~110.0%.The proposed method was successfully applied to the determination of xanthopterin and isoxanthopterin in human urine.
