摘  要:目的探讨蜂胶黄酮对培养大鼠乳鼠心肌细胞内游离钙离子的影响及其作用机制。方法原代培养SD大鼠乳鼠心肌细胞,用钙敏荧光探针Fura-2/AM负载染色后,采用阳离子测定系统,检测给药前后心肌细胞游离钙离子浓度[Ca2+]i及心肌细胞钙瞬变。结果 (1)静息状态下,心肌细胞[Ca2+]i为(83.3±11.7)nmol/L(x±s,n=6),终质量浓度分别为0.2、1、5μg/mL蜂胶黄酮对静息[Ca2+]i无影响。(2)KClI、so能明显升高[Ca2+]i(P<0.01),预孵蜂胶黄酮能抑制KCl及Iso引起的[Ca2+]i增高(P<0.01);(3)蜂胶黄酮能降低心肌细胞钙瞬变峰值,抑制钙瞬变频率,对KCl、异丙肾上腺素诱导心肌细胞钙瞬变变化具有抑制作用。结论蜂胶黄酮可抑制心肌细胞内钙超载,其作用机制可能与影响钙通道及细胞内钙释放有关。AIM To measure free Ca^2+ concentration of cultured cardiac cells in neonatal rats and observe the protective effect of flavone isolated from propolis on i.METHODS Two-day-old SD rats neonatal cardiac myocytes were cultured.Fura-2/AM was loaded as calcium probe.The change of i and i-oscillation was observed by cation meaturement system before and after various concentration of propolis flavone were added.RESULTS(1) the value of Ca^2+ was(83.3±11.7) nmol/L in the resting cardiac cells,no significant effect of flavone isolated from propolis 0.2,1,5 μg/mL on resting i was found.(2) KCl and Isoprenaline triggered a sustained rise in i,pretreatment with flavone isolated from propolis inhibites the elevation of i induced by KCl and Isoprenaline.(3)flavone isolated from propolis caused a decrease in the peak of Ca^2+ transient and inhibite the stimulatory effect of KCl and Isoprenaline.CONCLUSION Flavone isolated from propolis inhibite the i overload induced by KCl,Isoprenaline.One of the mechanisms seems to be that it reduces Ca^2+ influx via the L-type voltage-dependent calcium channels and release from the sarcoplasmic reticulum.
