摘 要:目的探讨黄芪对人脐静脉内皮细胞(VEC-304)增殖及血管内皮细胞生长因子(VEGF)表达的影响。方法以不同浓度的葡萄糖、黄芪、不同浓度黄芪联合24hIC50葡萄糖作用于VEC-304 24h,观察细胞生长状态变化。MTT法检测细胞生长率。应用RT-PCR检测24hIC50葡萄糖组、黄芪联合24hIC50葡萄糖组VEGF的mRNA表达;Western blot检测葡萄糖组、黄芪组和黄芪联合24hIC50葡萄糖组VEGF蛋白水平。结果 (1)不同浓度的葡萄糖和低浓度黄芪(50mg/L)联合24hIC50葡萄糖对细胞起抑制作用,可显著抑制VEC-304的生长;高浓度黄芪及高浓度黄芪(≥300mg/L)联合24hIC50葡萄糖对细胞有增殖作用。(2)葡萄糖组VEGF mRNA和蛋白表达下调,高浓度黄芪联合24hIC50葡萄糖组VEGF mRNA和蛋白表达正常。结论高浓度葡萄糖抑制VEC-304增值,高浓度黄芪促进VEC-304增值,黄芪可保护高浓度葡萄糖所致的血管损伤。Objective To investigate the of protective effects of radix astragali on the multiplication of traumatic vascular endothelial cells and the expression of vascular endothelial growth factor exposed.Methods Vascular endothelial cells,VEC-304 were treated with various concentrations of glucose,radix astragali and the combination of gluose and radix astragali.Morphological changes of VEC-304 were observed in 3 groups after 24h in vitro.The growth ratio was measured by MTT assay.The expression of VEGF mRNA in VEC-304 was tested by RT-PCR in two groups(24hIC50 glucose group,radix astragali group combined with 24hIC50 glucose).The expression of protein VEGF in three groups(glucose group,radix astragali group and the combination group of radix astragali and 24hIC50 glucose) were examined by Western blot respectively.Results(1) there were significant inhibitory effects on The VEC-304 treated with different concentrations glucose and the combination of light concentration radix astragali and 24hIC50 gluose.compared with control group,the growth of VEC-304 could be promoted by the high concentration radix astragali and the combination of high concentration radix astragali and 24hIC50 gluose.(2)The expression of VEGF mRNA and protein were depressed in glucose group,but it was at normal level in group of high concentration radix astragali combined with 24hIC50 glucose.Conclusion The growth of vessel endothelial cells could be inhibited by high concentration of glucose.High concentration radix astragali could promote the growth of vessel endothelial cells.radix astragali had the protective effects on the traumatic vascular endothelial cells exposed to high level of gluose.
