中药消痈溃得康对乙酸胃溃疡模型大鼠PGE_2及EGF含量的影响

摘　　要：目的:研究中药消痈溃得康对乙酸胃溃疡模型大鼠的治疗作用及其对大鼠前列腺素E2(PGE2)、表皮生长因子(EGF)含量的影响。方法:将大鼠随机分为正常组、假手术组、胃溃疡模型组、消痈溃得康治疗组及奥美拉唑治疗组。局部注射乙酸法制备胃溃疡模型,术后第2天开始给药,假手术组和胃溃疡模型组给纯水,术后第12天取血清及溃疡局部胃组织,酶联免疫法(ELISA)检测PGE2、EGF含量。结果:模型组大鼠胃组织及血清PGE2含量、胃组织EGF含量低于正常组和假手术组,差异有统计学意义(P<0.01);消痈溃得康治疗组和奥美拉唑治疗组大鼠胃组织及血清PGE2含量、胃组织EGF含量明显高于胃溃疡模型组,差异有统计学意义(P<0.01)。结论:消痈溃得康治疗胃溃疡的机制之一可能是通过促进胃、十二指肠黏膜合成、分泌、释放PGE2,增加黏膜上皮EGF的含量,维护胃黏膜的完整性,从而增强胃黏膜防御、修复功能。Objective： To study the effect of Xiaoyongkuidekang on expressions of PGE2,EGF in Rat Ulcer Model.Methods： Rats were divided into 5 groups randomly： normal group,sham group,ulcer model group,Xiaoyongkuidekang group and Omeprazole group.Acetic acid was applied to a certain part of the gastric mucosa to establish the ulcer model.The Xiaoyongkuidekang treatment group had been administrated with drugs since the 2nd day after the operation and the serum was taken on the 12th day,the content of PGE2 was measured by ELISA method.A part of the tissue taken from the focal area was pulped and the PGE2,EGF content was measured by ELISA method.Results： The PGE2 content of serum and the PGE2,EGF content of gastric tissue in ulcer model group was lower than that in the control group or in the sham group（P0.01）,The PGE2 content of serum and the PGE2,EGF content of gastric tissue were much higher in the Xiaoyongkuidekang group and the Omeprazole group than those in the ulcer model group（P0.01）.Conclusion： The molecular mechanism of stimulating the healing of gastric ulcer by applying Xiaoyongkuidekan may be related to increasing PGE2,EGF content.It is one of the most important defensive factors for protecting gastric mucosa.
