逆针灸关元穴对自然更年期大鼠下丘脑及腹部脂肪胰岛素受体表达的影响

摘　　要：目的:观察逆针灸关元穴对自然更年期大鼠胰岛素受体表达的影响。方法:经阴道筛查出动情周期紊乱的10月龄SD雌性大鼠60只,随机分为自然10、12、14、16月龄组、逆灸12、14、16月龄组和逆针12、14、16月龄组。另选3.5月龄健康SD雌性大鼠6只为年轻对照组。逆灸、逆针各组从大鼠10月龄开始,每周灸或针关元穴2次,共8周。采用免疫组化法检测下丘脑腹内侧核及腹部脂肪胰岛素受体的表达。结果:与年轻对照组相比,自然更年期大鼠下丘脑胰岛素受体表达自然10、12月龄组升高,自然14、16月龄组降低(P<0.05或P<0.01);腹部脂肪胰岛素受体表达自然10月龄组明显降低(P<0.01),自然12月龄组有升高趋势,自然14、16月龄明显降低(P<0.01)。逆灸、逆针处理后,大鼠下丘脑胰岛素受体表达与自然同月龄组比较,逆灸12月龄组显著升高(P<0.01),逆针14月龄组阳性细胞数、总灰度和逆灸16月龄组平均灰度、总灰度显著升高(P<0.05或P<0.01);大鼠腹部脂肪胰岛素受体表达逆灸12月龄组阳性细胞数、总灰度,逆针12月龄组、逆灸14月龄组阳性细胞数、平均灰度、总灰度和逆针16月龄组阳性细胞数变化具有显著性差异(P<0.05或P<0.01)。结论:逆针灸关元穴对更年期大鼠中枢及外周胰岛素受体表达具有良性调节作用,从而改善更年期机体的胰岛素抵抗状态,调节脂质代谢紊乱。Objective：To investigate the influence of preventive acupuncture and preventive moxibustion at Guanyuan（CV4） point on the expression of insulin receptor in hypothalamus and abdominal fat of natural climacteric model rats.Methods：A total of 60 female SD rats were randomized into 10th,12th,14th,16th month control groups,12th,14th,16th month preventive moxibustion groups and 12th,14th,16th month preventive acupuncture groups with 6 rats in each group.In addition,other 6 female SD rats aged 3.5th month were used as young control group.In preventive acupuncture and preventive moxibustion groups,Guanyuan was punctured with the needle retained for 20 min,and given with moxibustion for one moxa-cone respectively from aged 10th month.The treatment was conducted twice every week,8 weeks altogether.The expression of insulin receptor in hypothalamus and abdominal fat was observed by immunohistochemical method.Results： As the growing of month,the expression of insulin receptor in hypothalamus during early menopause increased and then decreased,the expression of insulin receptor in abdominal adipose tissues of meopausal rats decreased at the beginning,increased in the middle stage and decreased in the end.With preventive acupuncture and preventive moxibustion at Guanyuan point,the expression of insulin receptor in hypothalamus and abdominal fat presented an adverse trend compared to the same month control groups in most of the groups.Conclusion：Preventive acupuncture and preventive moxibustion at Guan-Yuan point had an benign regulation on the expression of insulin receptor in hypothalamus and abdominal adipose tissues,which might be one of the mechanism of preventing effectively dyslipidosis.
