参附益心颗粒对慢性心力衰竭大鼠心肌重塑的影响

摘　　要：目的:观察参附益心颗粒对慢性心力衰竭大鼠心肌重塑的影响。方法:随机将150只雄性SD大鼠分为造模组130只与假手术组20只,造模组结扎左冠状动脉前降支,假手术组手术方法同上,仅在冠状动脉前降支下穿线不结扎。造模8周后,经超声测定大鼠左室射血分数%(LVEF),<50%者随机分为5组,每组10只,分别为卡托普利组、氯沙坦组、参附益心颗粒高剂量组、参附益心颗粒低剂量组、模型组,另选假手术组大鼠10只。假手术组及模型组以纯净水灌胃,药物干预组将药物溶于纯净水中灌胃给药,每天1次,4周后查LVEF,称重后,处死测左室重量指数(LVMI)以及取组织观察病理组织学和心肌细胞超微结构。结果:参附益心颗粒有明显改善LVEF作用,明显地降低LVMI,可以改善心衰心肌病理形态和心肌超微结构的损伤而影响重塑,并且具有和西药对照组相近似的干预作用。结论:参附益心颗粒可延缓或逆转心肌重塑的作用。Objective： The present study was undertaken to observe the effect of the Shenfuyixin Granule on chronic heart failure rats myocardial remodeling.Methods： 150 heathly male Wister rats were randomly divided into sham operation group（n=20） and model group（n=130）.In model group the left anterior descending coronary artery was occluded,in sham operation group the sutures were passed under the left anterior descending coronary artery without ligation.After 8 weeks the modeling group were randomly assigned to 5 groups（n=10 in each group） based on the value of left ventricle ejection fraction（LVEF）50% obtained by Color Doppler Ultrasound,such as high dose of Shenfuyixin Granule,low dose of Shenfuyixin Granule,captopril group,losartan group,model group.All the medicine was given through gavage once a day for 4 weeks,the rats in model group and sham operation group（n=10） were administered lavage with equivalent sodium chloride once a day for 4 weeks.After 4 weeks,the LVEF was valued with Color Doppler Ultrasound,weighs were measured.Rats were killed.The left ventricular mass index（LVMI） was checked.Tissues were collected to observe pathological histology and myocardial ultra structure.Results： Shenfuyixin Granule can improve the LVEF obviously,low the LVMI and improve the myocardial pathomorphology and ultra structure of chronic heart failure in rats,which were equal to captopril group and losartan group.Conclusion： Shenfuyixin Granule can inhibit or reverse the cardiac remodeling.
