骨化三醇对血管紧张素Ⅱ诱导的大鼠肾小管上皮细胞转分化的影响

摘  要:目的骨化三醇在血管紧张素Ⅱ(AngⅡ)诱导的大鼠肾小管上皮细胞(NRK52E)表型转化中的作用。方法体外传代培养NRK52E后按以下分组加入不同干预因素,骨化三醇终浓度为10-6mmol/L,AngⅡ终浓度为10-6mmol/L,①对照组:NRK52E培养液中不加入干预因素;②AngⅡ组:NRK52E培养液中加入AngⅡ;③骨化三醇组:NRK52E培养液中加入骨化三醇;④AngⅡ+骨化三醇组:NRK52E培养液中加入AngⅡ和骨化三醇。经AngⅡ和骨化三醇干预12、24、48h后,应用细胞免疫化学染色法检测E-钙粘蛋白(E-cadherin)和α-肌动蛋白(α-SMA)的表达,应用酶联免疫吸附试验(ELISA)法检测Ⅰ型胶原(ColⅠ)和纤维粘连蛋白(FN)的表达。结果①与对照组比较,AngⅡ作用48h后,E-cadherin的表达减弱(P<0.05),α-SMA的表达显著增强(P<0.05),同时加入AngⅡ和骨化三醇后,与AngⅡ组比较,E-cadherin的表达增强(P<0.05),α-SMA的表达减弱(P<0.05);②AngⅡ作用48h后,细胞上清液细胞外基质成分FN、ColⅠ含量在AngⅡ组较对照组明显升高(P<0.05),同时加入骨化三醇和AngⅡ后,与AngⅡ组比较,FN、ColⅠ分泌有明显减少(P<0.05)。结论①骨化三醇能抑制AngⅡ诱导的NRK52E细胞的表型的转化;②骨化三醇能减少血管紧张素Ⅱ诱导的NRK52E细胞外基质FN、ColⅠ的分泌。Objective To explore the influence of calcitriol on angiotensionⅡ induced rat's tubularepithelial cell Epithelial-myofibroblast transition.Methods Experiments were performed after the cells had been cultured in the serum-free DMEM for 24 hours.The cells were divided into four groups according to the different intervention factors,final concentration of calcitriol was 10-6mmol/L,Ang Ⅱwas 10-6mmol/L.①Control group:the NRK52E were cultured without any treatment;②Ang Ⅱgroup:the NRK52E were stimulated by Ang Ⅱ;③Calcitriol group:the NRK52E were stimulated by calcitriol;④Ang Ⅱ+ calcitriol group:the NRK52E were co-stimulated by Ang Ⅱ and calcitriol.The cultured NRK52E cells,treated with Ang Ⅱand calcitriol for 12,24,48 hours,detected the expressions of E-cadherin and α-SMA by immunocyto-chemistry method,detected ColⅠand FN by ELISA.Results ①Treat with AngⅡ48 hours,expressions of E-cadherin decreased(P0.05),but α-SMA increased significantly(P0.05);②Content of FN and ColⅠ:Content of FN and Col Ⅰin the AngⅡ group was more obviously advanced than that of control group(P0.05),and calcitriol significantly inhibited the secretion of FN and ColⅠ induced by Ang Ⅱ(P0.05).Conclusion Calcitriol can inhibit Ang Ⅱ-induced the expression of α-SMA of NRK52E,increase the expression of E-cadherin.
