抗人白蛋白单克隆抗体制备与尿微量白蛋白半定量金标试纸研制

摘　　要：目的制备抗人白蛋白单克隆抗体和尿微量白蛋白（microalbuminuria，MCA）半定量试纸，作MAC检测，与传统检测方法比较其灵敏度和特异性。方法在制备抗人白蛋白单克隆抗体的基础上，采用人白蛋白作为包被抗原，应用胶体金免疫层析技术竞争抑制法原理制备出人尿微量白蛋白半定量检测试纸。用所研制的试纸与市售尿白蛋白ELISA诊断试剂平行检测50例正常健康成人晨尿样品和56例临床样本。结果用所研制的试纸与ELISA诊断试纸二者符合率达100％。结论本研制试纸完全可以满足临床检测要求，且操作简单、灵敏度高、特异性强、稳定性好，具有广泛的应用前景及广阔的发展空间。Objective To manufacture anti - human monoclonal antibody and strips detecting human microalbuminuria basing on colloid gold immunochromatography technology, for MAC detection, and to compare it with conventiona methods. Methods Based on the manufacture of anti - human monoclonal antibody and the theory of competitive inhibition, using human albumin as coating antigen and McAb against human albumin we developed labelling colloid gold. The strips and ELISA kid for human urine albumin were also used in parallel detecting urine samples from 50 normal persons and 56 clinic patients. Results The strips and ELISA kid matched in 100%. Conclusions This indicates that the strips were fit for clinic detection. Plus the characters of easy to operate, high sensitivity and specificity, the strips have a wide application prospect and an infinite potential for future development.
