摘 要:建立一种对天然绿色棉色素进行相对定量分析的方法,并在此方法的基础上对超声波萃取与普通浸提萃取的效果进行定量分析,通过紫外分光光度法研究了天然绿色棉色素乙醇溶液的紫外光谱性质,分析得出360 nm处的吸收值与色素浓度呈线性关系,R2值达到0.999 2。利用提出的相对定量方法研究了温度、时间、溶剂体积对萃取效果的影响,并与普通浸提进行了对比,结果发现在同等条件下超声波辅助萃取效率明显高于普通浸提,超声波辅助萃取天然绿色棉色素20 min达到普通浸提5 h的萃取效果,可缩短色素分析实验的周期。研究发现天然绿色棉色素溶液的颜色随着pH值的增加而加深,具有很强的pH值依赖性。The goal of this research work is to establish a method for the relative quantitative analysis on the pigment in natural green cotton,and on this basis,the paper made a quantitative analysis on the effects obtained by ultrasonic-assisted extraction and normal extraction separately.The UV spectral character of the pigment of natural green cotton in ethanol solution by UV spectrophotometry was studied.It was found that there is a linear relationship between the absorption value and pigment concentration at 360 nm,R2=0.999 2.The established relative quantitative analysis method was used to investigate the influence of temperature,time and the volume of the solution on the extraction,and comparison results showed that the efficiency of ultrasonic-assisted extraction was significantly higher than that of normal extraction at the same conditions.The efficiency of ultrasonic-assisted extraction of natural green cotton pigment for 20 min was equivalent to that of ordinary extraction for 5 hours,thus shortening the test cycle of the pigment.Research found that the color of pigment solution of natural green cotton darkens with the increase of pH value,and has intensive pH value dependence.
