实验性大鼠羊水栓塞角蛋白16表达的变化及意义

摘　　要：目的本实验通过复制羊水栓塞动物模型,研究不同性质、不同剂量羊水入血后角蛋白16表达的变化,探讨角蛋白16在羊水栓塞法医学鉴定中的意义。方法选用待产的Wistar雌性大鼠36只,随机分为三组,每组12只,分别为羊水原液组(AF)、羊水胎粪混合液组(MAF)、对照组(NS),每组各4只以1.25ml/kg、2.5ml/kg、3.75ml/kg不同剂量从舌下静脉分别注射羊水原液、羊水胎粪混合液、生理盐水,待大鼠死亡后取肺组织,进行HE染色及免疫组化染色,镜下观察角蛋白16表达的变化。结果对照组角蛋白16表达呈阴性反应,AF组和MAF组角蛋白16表达呈阳性反应,且随着羊水入血剂量的增加,角蛋白16表达阳性数增加,且染色更深,两组之间角蛋白表达无显著差异。结论角蛋白16的表达与羊水入血量有关,与羊水胎粪污染程度相关性不大。Objective To study the significance of keratin 16 in forensic,replicate the animal models of amniotic fluid embolism after injecting the different nature and doses amniotic fluid.Methods The Wistar female rats were randomly divided into 3 groups,there was the amniotic liquid group（AF）,the mixture of amniotic fluid meconium group（MAF） and the control group（NS）,and different doses of amniotic fluid were injected.The expression of keratin 16 was observed by the HE staining and immunohistochemical staining after the rats dying.Results The expression of cytokeratin 16 was negative in the control group,was positive in AF group and MAF group,and the numbers of keratin 16 expression increased when increasing amniotic fluid.Conclusions The expression of cytokeratin 16 relevants with the amniotic fluid into the blood,not relevant with the level of meconium-stained amniotic fluid.
