糖尿病和卵巢去势对大鼠NO、TGF-β_1及骨密度的影响及相互作用

摘  要:目的探讨糖尿病及卵巢去势对一氧化氮(NO)、转化生长因子β1(TGF-β1)、骨密度的影响及其相互作用。方法雌性Wistar大鼠160只,随机分为4组:正常对照组(A组),糖尿病模型组(B组,链脲佐菌素诱导制备),单纯去势模型组(C组,切除双侧卵巢),糖尿病并发去势模型组(D组,制成糖尿病模型后切除双侧卵巢),于造模后4,8,12,16 w,进行骨密度检测,并应用放免法检测血清雌激素水平,硝酸还原酶法检测血清NO水平,ELISA法检测血清TGF-β1水平,免疫组织化学法检测骨中TGF-β1水平。结果糖尿病及卵巢去势均可使血清NO水平逐渐降低(P<0.05),两者同时存在时,即糖尿病并发卵巢去势模型,血清NO水平降低更为明显。糖尿病可使血清TGF-β1水平升高(P<0.05),卵巢去势可使血清TGF-β1水平降低(P<0.05),但两者同时存在时血清TGF-β1变化不明显(P>0.05)。糖尿病及卵巢去势可使大鼠股骨骨密度降低(P<0.05),两因素同时存在时,股骨骨密度降低更明显。但在上述指标变化中,统计分析两者无明显的协同或拮抗作用,我们推测两因素间存在有叠加作用。结论糖尿病与卵巢去势同时存在,骨密度降低幅度更大,可出现更严重的骨质疏松。Objective To study the combined effects of estrogen deficiency and diabetes on nitric oxide(NO),TGF-β1 and bone mineral density(BMD) in rats.Methods The rats were randomly divided into 4 groups: sham-operated(A),a streptozotocininduced diabetes(B),an ovariectomized(OVX,C),and a combined OVX and streptozotocin-induced diabetes groups(D).After 4,8,12 w and 16 w,the BMD was measured,and serum estrogen levels was measured by radioimmunoassay method,serum NO by nitrate reduction method,serum TGF-β1 levels by ELISA method,the content of TGF-β1 in the bone tissues by the immunohistochemical method.Results Compared with group A,the level of serum NO and the BMD of femur became lower in group B,C and D(P〈0.05).And in group D,they became much lower.The serum TGF-β1 became higher in group B(P〈0.05),and it became lower in group C(P〈0.05),but there was no significant difference between goup D and group A(P〉0.05).And there was no significant interaction between diabetes and estrogen deficiency on NO,TGF-β1 and BMD.Conclusions Combined effect of diabetes and estrogen deficiency are not synergistic or counteractive but additive.
