摘  要:目的探讨T-bet,GATA-3和RORC在过敏性紫癜患者发病中的作用机制。方法采用SYBR GreenI实时荧光定量PCR法检测32例过敏性紫癜患者和15例正常人外周血单核细胞中T-bet,GATA-3和RORC mRNA的表达水平。结果过敏性紫癜患者与正常人T-bet基因ΔCt平均秩次值分别为27.70和16.10,患者组T-betmRNA表达水平低于对照组;过敏性紫癜患者与正常人GATA-3基因ΔCt平均秩次值分别为18.25和36.27,患者组GATA-3 mRNA表达水平高于对照组,过敏性紫癜患者与正常人RORC基因ΔCt值分别为2.00±0.46与3.29±1.57,患者组RORC mRNA表达水平高于对照组,差异均有统计学意义(P均<0.05)。结论转录因子T-bet,GATA-3和RORC可能在过敏性紫癜发病机制中具有重要作用。Objective To explore the roles of transcription factors T-bet, GATA-3 and RORC in the pathogenesis of lISP. Method SYBR Green 1 real-time tluoresccnce quantitative PCR technique was appled to detect the expressions of T-bet mRNA, GATA-3 mRNA and RORC mRNA in peripheral bh,od mononuclear cells ( PB- MCs) 32 HSP patients and 15 healthy persons'. Results The mean ranks of gene T-bet ACt both in the HSP group and the control group were 27.70 and 16.10, the expression level of T-bet mRNA in the HSP group was lower than that in the control group ( P 〈 0.05 ). The mean ranks of gene GATA-3 ACt in the HSP group and the control group were 18.25 and 36.27, the expression level of GATA-3 mRNA in the HSP group was higher than that in the control group (P 〈0.05). Tbe gent RORC ACt values in the HSP group and the control group were 2.00 ±0.46 and 3.29 ± 1.57, the expression level of RORC mRNA in the HSP group was higher than that in the control group (all P 〈 0.05). Conclusion Transcriplion factors T-bet, GATA- 3 and RORC may play important roles in the pathogenesis of HSP.
