摘 要:目的:从基因水平调查遵义地区汉族人群与流行性出血热(EHF)相关的人类白细胞抗原(HLA)-A*31、B*40、B*58、DRB1*16等位基因分布状况。方法:应用聚合酶链反应-序列特异性引物(PCR-SSP)方法对遵义汉族200名健康个体进行HLA-A、B、DR位点基因分型,筛选出HLA-A*31、B*40、B*58、DRB1*16阳性标本,进一步做高分辨基因亚型分型,进而对遵义汉族人群的这几组等位基因与其他不同人群的分布情况做比较。结果:遵义地区汉族HLA-A*31、B*58、DRB1*16基因各检出1种亚型,HLA-B*40共检出4种等位基因亚型,其中B*4001、B*4002、B*4006为常见亚型,B*4063为罕见基因亚型。遵义汉族人群所检出的等位基因频率大部分与我国其他地区的相接近,而与日本人比较差异有统计学意义(P<0.01)。结论:本研究获得了遵义汉族人群与EHF相关的HLA等位基因分布特征,为HLA与疾病相关性及本地区群体遗传学等方面的研究提供了重要依据。Objective: To investigate the allelic polymorphism of human leukocyte antigen (HLA)-A*31, B*40, B*58 and DRB 1 * 16 group, which are associated with the epidemic hemorrhagic fever(EHF), in Chinese Han population of Zunyi area. Methods: Polymerase chain reaction-sequence specific primers (PCR-SSP) method was applied to determine HLA-A, B and DR genotypes of 200 unrelated healthy individuals in Zunyi area. The positive samples of HLA-A*31, B*40, B*58 and DRBl*16 were selected and analyzed at the high resolution level. The allele frequency distribution in this population was compared with that in other populations. Results: Only 1 subtypes of HLA-A*31 allele, HLA-B*58 allele and HLA-DRBI* 16 were detected respectively in Chinese Han population of Zunyi area. Four HLA-B*40 alleles were detected, in which B* 4001, B*4002 and B*4006 were the common alleles, but HLA-B*4063 allele was a rare subtype. There was a close relation- ship in the HLA allele frequencies between Zunyi Han population and other Chinese Han populations, but there was a significant difference when compared with that of Japanese population (P 〈 0.01). Conclusion: The results showed the characteristics of the HLA alleles related to EHF distribution in Chinese Hart population of Zunyi area, and provided more insights into the studies and applications association with HLA genes
