摘 要:目的:以老龄大鼠为研究对象,观察左旋丁苯酞(L-3-n-Butylphthalide,1-NBP)对脑低灌注大鼠学习记忆的改善作用,并研究其皮质及海马β-淀粉样肽(Aβ1-42)水平变化。方法:永久性结扎老龄大鼠双侧颈总动脉3个月,制备脑低灌注模型。实验分4组:假手术组、溶剂对照组、30 mg·kg^(-1)-NBP和120 mg·kg^(-1)-NBP组。1-NBP组连续给予1-NBP 45 d后,用Morris水迷宫检测大鼠的空间学习和记忆以及保存记忆的能力。同时,取大鼠脑皮质及海马组织,用ELISA法检测Aβ1-42含量,以探讨1-NBP改善认识功能作用机制。结果:1-NBP可以显著缩短脑低灌注大鼠找到安全台的潜伏期,并明显增加脑低灌注大鼠在目标象限活动时间的百分比,明显降低大鼠皮质及海马Aβ1-42含量。结论:1-NβP能改善脑低灌注大鼠的学习记忆缺陷,减少皮质及海马Aβ1-42的水平,这可能是改善脑低灌注大鼠认知功能的重要机制之一。Objective: To observe the improving effects of L-3-n-butylphthalide (1-NBP) on learning and memory of cerebral hypoperfused aged rats and investigate the biochemical mechanism. Method: The cerebral hypoperfused rat model was made by ligating bilateral common carotid arteries of rats for three months. The rats were randomly divided into four groups, namely sham operation group,solvent control group,1-NBP group with dosage of 30 mg·kg^-1 and 1-NBP group with dosage of 120 mg·kg^-1. The two 1-NBP groups were administered 1 -NBP for 45 clays consecutively. The spatial learning and memory of rats were measured by Morris water maze and the β-amyloid peptide content in rat brain tissue was detected by ELISA method in order to judge the mechanism of improving cognitive function of 1-NBP. Result: In the water maze positioning navigation trial, 1-NBP could significantly reduce the latency of finding safe platform of cerebral hypoperfused rats. In spatial exploring trial, 1-NBP could increase the time percentages spent in the platformquadrant of cerebral hypoperfused rats. Meanwhile, the contents of cortex and hippocampus Aβ1-42 were decreased significantly. Conclusion: 1-NBP can improve learning and memory deficits and reduce the contents of cortex and hippocampus Aβ1-42 of cerebral hypoperfused rats to protect neurons,which may be one of the important mechanisms of improving cognitive function.
