摘 要:目的探讨燃煤型氟中毒对雄性大鼠生殖内分泌激素的影响。方法将105只雄性SD大鼠随机分为7组:对照组、低氟组(含氟20 mg/kg)、中氟组(含氟40 mg/kg)、高氟组(含氟60 mg/kg)、低氟加营养组、中氟加营养组、高氟加营养组。各染毒组喂饲含不同比例的燃煤型氟中毒病区煤烘玉米的饲料,复制燃煤型氟中毒动物模型。分别于染毒90、120和180 d处死动物,查看氟斑牙,测尿氟,检测大鼠睾丸内睾酮,血清T、E2、LH、FSH、GnRH。结果与对照组比较,各染毒组大鼠血清T、E2水平随染毒时间延长和染氟剂量增加逐渐降低[如高氟组,染氟90和180 d,血清车载睾酮(821.77±13.21)ng/dL vs(140.80±15.58)ng/dL,睾丸内睾酮(3 248.62±20.69)ng/dL vs(984.63±7.39)ng/dL](P<0.05);LH、FSH水平随染毒时间延长和染毒剂量增加逐渐增高;GnRH水平随染毒时间延长逐渐降低,同一时间点,染毒组之间,随染氟剂量增加变化逐渐加大。相同染氟剂量加营养组性激素水平均有所改善。结论燃煤型氟中毒对雄性大鼠生殖内分泌有明显的影响,并随染氟剂量增加和染毒时间延长而加重,饮食营养可改善其影响。Objective To study the effects of fluorosis of coal burning on reproductive endocrinology of male rats.Methods SD rats were randomly assorted into 7 groups: control group,low-dose fluoride group,middle-dose fluoride group,high-dose fluoride group,low-dose fluoride added nutrition group,middle-dose fluoride added nutrition group and high-dose fluoride added nutrition group.The exposed groups were fed with the fodder with different proportions of corn smoke dried by burning coal coming from endemic fluorosis areas,to establish the animal model of coal-burning fluorosis.All rats were killed on day 90th,120th,180th after treatment,and examinations of dental fluorosis,fluorine in urine,the concentrations of intratesticular testosterone(T),estradiol(E2),luteinizing hormone(LH),follicle stimulating hormone(FSH),Gonadotropin-Releasing Hormone(GnRH) in serum were carried out.Results Compared with the control groups,the level of T,E2 declined significantly with the increasing fluoride dose and the longer exposure time in exposed groups(P 0.05 or P 0.01),but the level of LH and FSH increased significantly with the increasing fluoride dose and the longer exposure time in exposed groups;the level of exposed groups'GnRH declined significantly with the longer exposure time but increased significantly with the increasing fluoride dose between exposed groups(P 0.05 or P 0.01).In the same dose of fluoride added nutrition groups sex hormone levels improved(P 0.05 or P 0.01).Conclusions Coal-burning fluorosis had a significant endocrine effect on reproductive endocrinology of male rats,and influence increased,with the increasing fluoride dose and prolonged exposure,reducing fluoride intake and improving nutritional status can ease the effects of coal-burning fluorosis on reproductive endocrinology of male rats.
