摘 要:通过生物活性检测发现Streptomyces tz92菌株嘧肽霉素发酵液中一种新的抗细菌生物活性物质,为了明确其适宜的分离纯化工艺,试验研究了利用大孔吸附树脂,硅胶柱层析和分子筛层析分离纯化该组分的条件和效果。试验结果显示,发酵液经过大孔树脂SP825L吸附50%甲醇洗脱、硅胶柱层析氯仿∶正丁醇∶甲醇∶乙酸=42∶2∶4∶2(体积比)洗脱、Sephadex LH-20 70%甲醇洗脱可以得到该组分纯品。A new anti-bacteria bio-active compound in Cytosinpeptidemycin fermentation broth was found by bioassay analysis.In order to develop a proper separation and purification technique for the compound,conditions and effects were investigated by silica gel column chromatography and molecular sieve chromatography with macroporous adsorptive resin.Results showed that pure compound could be obtained through absorption of SP825L and eluted by 50% methanol,silica gel column chromatography with Chloroform ∶ 1-butanol ∶ methanol ∶ acetic acid=42∶2∶4∶2(by vol.),Sephadex LH-20 70% methanol as mobile phases.
