摘  要:采用基因芯片技术检测了人脐带与骨髓两种不同来源间充质干细胞中miRNA的表达.结果表明:有26个miRNA差异表达,其中miR-29b、miR-20a、miR-31、miR-20b及miR-106a在人脐带间充质干细胞(UMSCs)中表达显著上调,差异倍数均高于3,miR-138则表达下调.在此基础上,采用实时荧光定量PCR技术对差异性显著的6个miRNA进行验证,检测结果一致.最后对特异性基因进行靶点预测,发现靶基因与mRNA转录、翻译以及细胞增殖、分裂、信号转导等功能相关.Expressions of miRNA in human mesenchymal stem cells derived from umbilical cord and bone marrow were detected by microarray.Results indicated that the expressions of a set of 26 miRNA were different in UMSCs and BMSCs.miR-29b,miR-20a,miR-31,miR-20b and miR-106a exhibited higher expressions in UMSCs,and fold changes were higher than 3.However,miR-138 demonstrated lower expression.Based on this study,6 miRNAs of particular interests were confirmed by real-time PCR,and the results coincidented with those of mircoarray.Target genes predictive were correlated with mRNA transcript,translation,and the functions of cell proliferation,division,and signal transduction.
