摘 要:目的:探讨丙戊酸镁对小鼠急性脑缺血再灌注损伤的预防作用及其机制。方法:取小鼠随机分为假手术组、模型组和丙戊酸镁高、低剂量(0.04、0.02mg·g-1)组,每组10只,灌胃给予相应药物,每日2次,连续14d,末次给药1h后对后3组小鼠建立急性脑缺血再灌注损伤模型,建模成功后检测各组小鼠脑指数和脑组织中丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,并观察大脑皮层和海马CA1区细胞形态的变化。结果:与模型组比较,丙戊酸镁高、低剂量组小鼠神经元损伤减轻,脑指数和MDA含量明显降低,GSH-Px活性明显升高(P<0.01或P<0.05),且丙戊酸镁高剂量组SOD活性明显升高(P<0.01)。结论:丙戊酸镁对小鼠急性脑缺血再灌注损伤具有预防作用,其机制可能与抗氧化作用有关。OBJECTIVE: To investigate the preventive effect of magnesium valprote on acute cerebral ischemia-reperfusion (IR) injury in mice and its mechanism. METHODS: Mice were randomly divided into sham operation group, model group, and magnesium valprote high-dose and low-dose groups (0.04, 0.02 mg·g^-1) with 10 mice in each group. Those groups were administrated with relevant drugs intragastrically twice a day for 14 days. 1 hour after the last administration, in the latter three groups acute cerebral IR injury models were established. After modeling, cerebral index, MDA content, activities of SOD and GSH-Px in cerebral tissue were determined, and the cell morphology in cerebral cortex and hippocampus CA1 were observed. RESULTS: Compared with model group, neural injury relieved in magnesium valprote high-dose and low-dose groups, cerebral index and MDA content decreased significantly, but activities of GSH-Px increased significantly (P〈0.01 or P〈0.05). Furthermore, SOD activity increased significantly in magnesium valprote high-dose group (P〈0.01). CONCLUSION: Magnesium valprote has preventive effect against acute cerebral IR injury, which is associated with anti-oxidation effect.
