Effects of modified Huanglian Jiedu decoction on nuclear factor-κB pathway in rats with multiple organ dysfunction syndrome

摘  要:目的探讨加味黄连解毒汤对多器官功能障碍综合征(MODS)大鼠核转录因子-κB(NF-κB)、一氧化氮(N0)等炎症介质通路的影响,以及对损伤器官的保护作用。方法将36只SD大鼠按随机数字表法分为对照组(6只)、模型组(15只)、中药组(15只)。通过腹腔注射酵母多糖法复制MODS大鼠模型。中药组灌胃加味黄连解毒汤25ml/kg;对照组、模型组则灌胃生理盐水,12h1次。于制膜12、24、48h观察外周血、小肠、心脏组织一氧化氮(N0)系统及NF-κB的变化。结果与对照组比较,模型组12h时血清NO(μmol/L)及诱生型一氧化氮合酶(iNOS,kU/L)即明显升高(NO:167.16±15.10比82.35±11.40,iNOS:10.20±1.20比5.48±1.16),结构型一氧化氮合酶(cNOS)明显减少(10.40±1.87比13.08±2.68,均P〈0.05);与模型组比较,中药组12h时NO(113.14±10.08)、iNOS(6.52±0.85)即明显降低,eNOS(13.63±1.46)明显升高(均P〈0.05)。模型组小肠、心脏组织N0和iNOS明显升高,eNOS明显降低;中药组N0和iN0s明显降低,eNOS明显升高(均P〈0.05)。对照组小肠偶见NF-κB表达阳性细胞,心脏组织中几乎无NF-κB表达;模型组小肠及心脏组织可见NF-κB阳性细胞;中药组小肠NF-κB阳性细胞数明显减少,心脏组织未见明显降低。结论加味黄连解毒汤能有效改善MODS大鼠器官功能,其机制可能是通过抑制NF-κB表达从而减轻促炎介质及iNOS过度活化。Objective To observe the protective effects of Huanglian Jiedu decoction (黄连解毒汤) and Dahuang (HLJDT) on organ function and structure and its effects on nuclear factor-κB (NF-κB) and nitric oxide (NO)/nitric oxide synthase (NOS) system in rats with multiple organ dysfunction syndrome (MODS). Methods Thirty-six rats were randomly divided into control (n= 5), model (n= 15) and HLJDT treatment (n= 15) groups. The animal models of MODS were induced by intra-peritoneal injection of zymosan and the modified HLJDT was given orally. NF-κB in blood plasma were detected, and the changes in NF-κB, NO/NOS system in peripheral blood, small intestine, heart, etc. were examined. Results Compare with the control group, the serum levels of NO (μmol/L) and induced type of nitric oxide synthase (iNOS, kU/L) were significantly increased (NO: 167.16±15.10 vs. 82.35±11.40, iNOS: 10. 20±1.20 vs. 5.48±1.16), while the serum level of constitutive nitric oxide synthase (cNOS) was obviously decreased (10. 40±1.87 vs. 13.08±2.68) in model group after 12 hours of observation (all P〈0.05). Compared with those in model group, after treatment with HLJDT for 12 hours, the levels of NO (113.14± 10. 08) and iNOS (6.52± 0. 85) were decreased markedly and the level of cNOS (13.63±1.46) in serum was increased remarkably (all P〈 0.05). In the model group, the levels of NO and NOS were significantly increased, and cNOS markedly decreased, while in Chinese medicine group, the levels of NO and cNOS were obviously lowered and cNOS remarkably elevated (all P〈0.05) in small intestine and heart tissues. In the control group, occasionally there were a few cells with positive expression of NF-κB in small intestine, while in the heart tissue, there was almost no expression of NF-κB in the model group, there were cells with positive expression of NF-κB in small intestine and heart tissues, while in the HLJDT group, the number of cells with positive
