摘 要:目的研究亚慢性低剂量2,3,7,8-四氯二苯-并-二噁英(2,3,7,8-tetrachlorodibenzo-p-dioxin,TCDD)染毒对SD雄性成年大鼠海马形态结构的影响。方法将32只1月龄SPF级Sprague-Dawley雄性大鼠随机分为4组,分别为对照(玉米油)组和低(2 ng/kg)、中(10 ng/kg)、高剂量(50 ng/kg)TCDD染毒组,每组8只。将相应体积的TCDD染毒溶液(或玉米油)涂抹在面包片(与动物标准饲料成分相当,排除二噁英污染)上,供动物自由食用,每天1次,连续染毒13周。取海马组织进行电镜超微结构观察,常规HE染色,海马CA1区体视学分析。结果 TCDD染毒大鼠海马各区细胞排列较对照组松散,锥体细胞超微结构的损伤以内质网同心圆样改变和线粒体空泡样变为主。各剂量TCDD染毒雄性大鼠海马CA1区锥体细胞的体积密度、表面积密度、平均体积、比表面积与对照组相比,差异均无统计学意义。结论亚慢性低剂量TCDD染毒可引起成年雄性大鼠海马区细胞排列松散,CA1区锥体细胞超微结构受到破坏。Objective To study the effects of low dose,long term exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)on morphology and ultrastrueture of hippoeampus in adult male rats. Methods Thirty-two one-month-old SPF male SD rats were randomly divided into 4 groups, 8 in each, and treated daily with TCDD at the doses of 2,10 and 50 ng/kg in the experimental groups,corn oil was given to the control. The rats in each group received daily corresponding volume of TCDD (or corn oil) daubed on a piece of bread with contents of matchable to the standard laboratory food, once a day ,for 13 weeks. After 13 weeks,pathological (ie. hematoxylin-eosin staining and stereology analysis on CA1 area) and uhrastruetural observation were conducted with transmission electron microscope on hippoeampus. Results Under light microscope ,the pyramidal cells' distribution in hippocampus of TCDD treated rats showed sparser than the control. The major damages in the pyramidal cells of TCDD treated groups were the mitochondrial vacuole and concentric ring-shaped endoplasmic reticulum. However,no statistical significance was found in volume density ,surface density ,mean volume and specific surface between experimental groups and control. Conclusion Subchronic TCDD exposure may results in sparse distribution of pyramidal cells in hippoeampus and damage the ultrastructure of pyramidal cells in CA1 area in adult male rats.
