摘  要:目的:建立聚合酶链反应偶联印迹杂交和酶联放大技术(即三联放大技术)对乙型肝炎病毒的检测方法,探索适用于血液筛查乙肝的核酸检测技术。方法:通过检测HBV定值标准血清,评估三联放大技术检测血清HBV DNA的灵敏度。然后,应用三联放大技术对乌鲁木齐市500人(次)血清学检测病毒标志物阴性的献血者进行HBV DNA检测。结果:建立三联放大技术HBV检测体系,该体系对90 IU/ml的HBV标准血清检出率为100%,对45 IU/ml的HBV标准血清检出率为75%。对500人(次)血清学检测病毒标志物阴性的献血者,未检出HBV DNA阳性。结论:三联放大技术对HBV检测灵敏度较高,可运用于血液筛查乙型肝炎病毒。Objective: To detect hepatitis B virus in serum by establishing the polymerase-chain reaction coupled with blot-hybridization and enzyme-linked technology (triple amplification technology). Discuss that nucleic acid amplification testing applies to detect HBV in blood donors screening. Methods: Triple amplification technology was used to detect HBV standard serum to evaluate the sensitivity of triple amplification technology detected HBV DNA in serum. 500 seronegative samples were detected by triple amplification technology for HBV DNA. Results: The HBV DNA detection rate of 90 IU/ml HBV standard serum was 100%, and the HBV DNA detection rate of 45 IU/ml HBV standard serum was 75% by triple amplification technology. 500 seronegative samples were detected that HBV DNA were negative. Conclusion: The results imply that triple amplification technology is a sensitive method to detect HBV and can be applied to detect HBV in blood donors screening.
