摘 要:[目的]研究聚酰胺颗粒分离纯化紫果西番莲总黄酮的工艺条件。[方法]采用紫外分光光度法测定紫果西番莲总黄酮的含量,通过考察静态、动态吸附试验及洗脱条件,得到聚酰胺颗粒分离纯化西番莲总黄酮科学合理的工艺条件。[结果]聚酰胺对紫果西番莲叶总黄酮有良好的吸附作用,静态吸附的饱和吸附量为23.14 mg/g。其动态吸附分离工艺条件:上样量30 ml,先用8 BV的水洗脱至无Molish反应,然后用12 BV的50%乙醇以 2BV/h的流速进行洗脱。[结论]聚酰胺颗粒对紫果西番莲叶总黄酮吸附选择性高,操作简单,可用于紫果西番莲的富集纯化。[Objective] To study the separation and purification of total flavonoids from Passiflora edulis with polyamide particles.[Method] The content of total flavonoids was determined with UV spectrophotometry,and the scientific and reasonable conditions for isolating and purifying total flavonoids from Passiflora edulis with polyamide particles were obtained through observing the static,dynamic adsorption and elution conditions.[Result] Polyamide could well adsorb the flavonoids in the leaves of Passiflora edulis,the saturated static adsorption capacity was 23.14 mg/g.The dynamic adsorption and isolation conditions were:30 ml Passiflora edulis extracts were eluted with 8 BV water until without Molish reaction,then eluted with 12 BV 50% ethanol at a rate of 2 BV/h.[Conclusion] The polyamide particles had high selectivity to the adsorption of total flavonoids from Passiflora edulis,which was easy to operate,therefore it could be used to the enrichment and purification of Passiflora edulis.
