豚鼠牙齿组织总RNA的提取研究

摘　　要：[目的]研究对RNA丰度值低、硬度大、处理困难的牙齿组织总RNA的提取和纯化技术。[方法]采集10只Hartley豚鼠的切齿和臼齿,分别对其进行了总RNA的提取和纯化,对所提纯的总RNA进行了琼脂糖凝胶电泳鉴定,以及浓度和纯度的测定。[结果]切齿和臼齿总RNA均可见明显的28S、18S和5S三条带,RNA浓度均大于100 ng/μl,A260/A280值均在1.7～2.0之间,A260/A230值在0.1～0.7之间,总RNA的提纯效果较好。[结论]为动物硬组织总RNA的提取提供了技术参考。[Objective] The extracting and purifying technique of total RNA from guinea pig teeth,which was with the characteristics of low abundance of RNA,high solid and treating hard,was studied.[Method] The total RNA,extracted from 10 guinea pigs’ incisor and molar,was purified and then identified with the technique of agarose gel electrophoresis and its concentration and purity was determined.[Results] There were clear three obvious bands（28s,18s and 5s）,which concentrations were beyond 100 ng/μL;values of A260/A280,between 1.7-2.0;A260/230,between 0.1-0.7,and this technology was successfully applied in extraction and purification of total RNA.[Conclusion] The reference for total RNA extracted from hard tissue of animal was provided.
