摘  要:目的检测小青龙汤对支气管哮喘小鼠磷酸酶基因(PTEN)和基质金属蛋白酶-9(MMP-9)的影响,探讨小青龙汤治疗支气管哮喘的机制。方法健康雌性BALB/c小鼠随机分成正常对照组、哮喘组、地塞米松组和小青龙汤组。以卵蛋白致敏法建立支气管哮喘小鼠模型,造模成功观察2周后,处死小鼠取肺组织,苏木精-伊红(H-E)染色观察病理组织变化,免疫组织化学法检测MMP-9和PTEN蛋白表达情况。结果哮喘组小鼠肺组织PTEN的表达量显著低于正常对照组(P<0.01),小青龙汤组和地塞米松组治疗后小鼠肺组织PTEN的表达量显著高于哮喘组(P值均<0.01)。哮喘组小鼠肺组织MMP-9的表达量显著高于正常对照组(P<0.01),小青龙汤组和地塞米松组治疗后小鼠肺组织MMP-9的表达量显著低于哮喘组(P值均<0.01)。结论小青龙汤能显著改善气道炎症。小青龙汤选择性上调PTEN、下调MMP-9表达,可能是其治疗哮喘的作用机制之一。Objective To investigate the effect of Xiaoqinglong decoction on the expression of phosphatase and tensin homolog (PTEN) and matrix metalloproteinases-9 (MMP-9) in the lung of asthma rats. Methods Health female BALB/c mice were randomly divided into control group, asthma group, dexamethasone group, and Xiaoqinglong decoction group. The mice models of asthma were established by egg albumin hypersensitivity. Two weeks after model establishment, these mice received corresponding treatments for 2 weeks, and then the mice were executed, their lungs were sampled immediately, and stained by H-E for morphological observation and by immunohistochemistry dye to observe the changes of PTEN and MMP-9 expression. Results The PTEN expression was significantly lower in asthma group than in normal control (P〈0.01), while that in Xiaoqinglong decoction group and dexamethasone group was significantly higher than that in asthma group (P〈0.01). Asthma group had significantly higher expression of MMP-9 than normal control (P〈 0.01), but Xiaoqinglong decoction group and dexamethasone group had significantly lower MMP-9 expression than asthma group (P〈0.01). Conclusion Xiaoqinglong decoction can attenuate bronchial inflammation, which might be due to up-regulation of PTEN and down-regulation of MMP-9. (Shanghai Med J, 2011, 34: 692-695)
