摘 要:目的研究脑源性神经营养因子对人胚胎脊髓神经干细胞生长和分化的影响。方法孕20周水囊引产胎儿标本经家属同意捐赠,分离培养脊髓神经干细胞,分为正常对照组、BDNF蛋白组、BDNF抗体封闭组,计数神经球数目并以MTT法检测细胞活力,免疫组织化学检测NeuN和GFAP的表达情况并进行计数。结果与对照组相比,BDNF蛋白组神经球数目或MTT值均无明显变化,BDNF抗体封闭组的神经球数目和MTT值有明显减少(P<0.05);BDNF蛋白组NeuN阳性细胞数明显多于对照组(P<0.05),GFAP阳性细胞数与对照组相比有明显减少(P<0.05),BDNF抗体封闭组NeuN或GFAP阳性细胞数均少于对照组(P<0.05)。结论外源性BDNF可促进人胚胎脊髓神经干细胞向神经元样细胞分化;内源性BDNF在人胚胎脊髓神经干细胞的增殖分化过程中发挥重要作用。Objective To investigate the brain-derived neurotrophic factor in human embryonic spinal cord neural stem cell growth and differentiation.Methods Human fetus aged 20 weeks induced by labor with water bag were donated by their parents.Human neural stem cells were isolated and cultured into control group,BDNF protein added group,BDNF antibody blocked group.Each group of cells were counted under a microscope and MTT assay were used to detect the cell viability.Immunohistochemistry were used to test the expression of NeuN and GFAP and then the positive cells were counted.Results Compared with the control group,the neurospheres′ number and MTT value in BDNF protein added group have no significant change,while the BDNF antibody blocked group′s neurospheres number and MTT value appeared significantly reduced(P0.05);the NeuN-positive cells in BDNF protein added group were obviously more than the control group(P0.05),while the GFAP-positive cells saw a significant decreas(P0.05).In the BDNF antibody blocked group,the number of either GFAP-positive cells or NeuN positive cells were less than the control group(P0.05).Conclusion Exogenous BDNF can promote the human embryonic spinal cord neural stem cells to differentiate into neuron-like cells;endogenous BDNF plays an important role in the proliferation and differentiation of human embryonic spinal cord neural stem cell.
