摘 要:目的探讨八味沉香散对脑缺血再灌注损伤大鼠海马CA1区神经元凋亡的影响。方法 Wistar大鼠随机分五组:常氧对照组、低氧组和八味沉香散大、中、小剂量组。将低氧组和八味沉香散组放入模拟海拔4 500m的低氧低压舱内。常氧对照组和低氧组以0.9%生理盐水灌胃,八味沉香散组对脑缺血再灌注损伤大鼠模型以八味沉香散灌胃。利用流式细胞仪检测海马神经元凋亡率、免疫组织化学法研究神经细胞Caspase-3的表达变化。结果八味沉香散高原组大鼠海马神经元凋亡率、Caspase-3免疫组化阳性细胞数显著高于低氧组;海马MDA含量低于低氧组,而SOD高于低氧组(P<0.05)。结论八味沉香散可降低脑缺血再灌注损伤大鼠海马CA1区神经元的凋亡率。Objective To explore the effect of Tibetan Bawei chenxiang powder on cerebral ischemia reperfusion injury rats with apoptosis neurons at hippocampal CAI area. Methods W/star rats were randomly divided into 5 groups for the normoxic control, hypoxic and Tibetan Bawei chenxiang powder scattered high, medium and low - dose groups. Rats in hypoxic and Tibetan Bawei chenxiang powder scattered high, medium and lowdose groups were placed into the low -voltage analog cabin altitude at 5 500 meters. Rats in normoxic control and the hypoxic groups were fed with 0.9% saline. Bawei chenxiang powder was irrigated to stomach. Using flow cytometry to detect apoptosis rate and hippocanlpal neurons. Immunohistochemistry methods were used to research Caspase -3 involving nerve cells expression. Results The hippocampal neurons apoptosis rate, Caspase -3 immunohistochemical positive cell counts in Bawei chenxiang powder group was significantly higher than that of low oxygen group. The MDA contents were lower than that of low oxygen group, and SOD higher than low oxygen group (P 〈 0.05 ). Conclusion Bawei chenxiang powder can reduce the cerebral ischemia reperfusion injury and apoptosis neurons rate at hippocampal CA1 area in rats.
