头针对急性脑缺血再灌注大鼠BrdU/nestin表达的实验研究

摘　　要：目的观察头针对局灶性脑缺血再灌注模型大鼠BrdU/nestin表达的影响,探讨头针治疗脑缺血的可能机制。方法将90只健康雌性Wistar大鼠随机分为假手术组、模型组和头针组,各组再根据缺血再灌注时间的不同,随机分为7d、14d、28d共9个亚组。采用大脑中动脉线栓法制备大脑中动脉闭塞(MCAO)再灌注模型,选取顶颞后斜线、顶颞前斜线,进针后接韩氏穴位神经刺激仪,每天治疗1次。对大鼠应用神经功能缺损评分(NSS),免疫荧光检测法观察脑缺血再灌注各时间点大鼠海马齿状回BrdU及BrdU/nestin含量。结果头针组与模型组各时相NSS组间比较,头针组显著低于模型组(P<0.05,P<0.01)。脑缺血再灌注侧海马齿状回BrdU的表达:模型组与假手术组比较均有显著性差异(P<0.01),头针组在各时间点上与模型组比较,均显著性升高(P<0.01),7 d达到峰值,14 d开始下降。脑缺血再灌注侧海马齿状回BrdU/nestin的表达:模型组与假手术组比较均有显著性差异(P<0.01),头针组在各时间点上与模型组比较,均显著性升高(P<0.05),以7 d最为明显(P<0.01)。结论头针可明显改善大鼠神经功能缺损状态,促进神经干细胞得再生,通过增加nestin的含量来促进急性脑缺血再灌注损伤过程中神经干细胞的修复,实现对缺血脑组织保护的目的。Objective To explore the possible mechanisms of Scalp-acupuncture（SA） in treating cerebral ischemia by observing the expression of BrdU/nestin in rats with focal cerebral ischemia reperfusion injury.Methods 90 Rat models with middle cerebral artery occlusion（MCAO） were randomized into sham operated group（without artery occlusion）,model group and SA group（the SA group was randomly subdivided into 7d,14d,18d groups according to the duration for reperfusion.The model group and the SA group were undergoing needles insertion along anterior oblique line of vertex-temporal and posterior oblique line of vertex-temporal,soon after when a Han＇s Electro-Acupuncture Apparatus was connected for treatment for once a day） and applied neurological severity score（NSS）,immunofluorescence assays were applied for detecting BrdU and BrdU/nestin contents of dentategyrus in each time point. Results（1） On NSS at each phase point：SA group demonstrated obvious decline in NSS at all time phases when compared with that of model group（P0.01）.On observation of BrdU at each phase,significantly differences were seen in SA group contrasting with that of model group and sham group（P0.01）.On observation of BrdU model group and SA group at each phase point,there were significant differences（P0.05）.There were significant differences in BrdU/nestin between scalp acupuncture group and model group（P0.05）,with the most significant difference was observed at the 7th day.Conclusions Scalp-acupuncture is protective for the brain against ischemia by obvious improvement on the deficit of neural function and the regeneration of the neural stem cells（NSCs）.In addition,SA increases nestin content for facilitating the repairment of the in-process ischemic injury NSCs.
