补肾益气活血方对胎儿生长受限胎鼠IGF-1、IGFBP-1及IGFBP-3表达的影响

摘　　要：目的观察补肾益气活血方对胎儿生长受限胎鼠肝脏IGF-1、IGFBP-1、IGFBP-3表达的影响,探讨补肾益气活血方治疗FGR的作用机制。方法采用被动吸烟法建立SD大鼠FGR模型,分为正常组、FGR组、补肾益气活血方治疗组(中药组)、精氨酸治疗组(精氨酸组)。采用免疫组织化学方法检测胎鼠肝脏IGF-1、IGFBP-1、IGFBP-3的表达。结果与FGR组比较,中药组胎鼠肝脏IGF-1、IGFBP-3的表达均明显升高(P<0.05),而IGFBP-1的表达明显降低(P<0.05),并且IGF-1的表达强度高于精氨酸组(P<0.05),各项指标与正常组比较差异无统计学意义(P>0.05)。结论补肾益气活血方可能通过调节IGF-I、IGFBP-1、IGFBP-3的表达,从而促进胎儿在宫内的生长发育。Objective To investigate the effect of Bushen Yiqi Huoxue（Reinforcing Kidney Replenishing qi and Promoting Blood Circulation） Decoction on the expression of IGF-1,IGFBP-1 and IGFBP-3 proteins in fetal liver of Fetal growth restriction（FGR） rat.Methods The smoke inhalation rat model was created.Thirty two pregnant Sprague-Dawley rats were randomly dividedly into four groups： control group,FGR group,group of FGR treated with Bushen Yiqi Huoxue Decoction（Chinese medicine group）,group of FGR treated with arginine（arginine group）.The expression of IGF-1,IGFBP-1 and IGFBP-3 proteins in fetal liver were analyzed by immunohistochemistry.Result Compared with FGR group,the expression of IGF-1 and IGFBP-3 in fetal liver of Chinese medicine group increased significantly（P0.05）,whereas the expression of IGFBP-1 decreased significantly（P0.05）;in addition,the expression of IGF-1 was higher than that of arginine group（P0.05）.The difference among Chinese medicine group and control group was insignificant（P0.05）.Conclusion The effect of Bushen Yiqi Huoxue Decoction on preventing and treating FGR may be attributed to regulating the expression of IGF-1,IGFBP-1 and IGFBP-3.
