摘  要:以‘富士’(Malus domestica‘Fuji’)、‘乔纳金’(M.‘Jonagold’)苹果试管苗为试材,研究了比久(丁酰肼,B9)、矮壮素(CCC)、多效唑(PP333)等生长延缓剂处理下苹果组培苗延缓生长以及恢复生长后SOD、POD活性和MDA含量的变化,以期为筛选苹果组培苗延缓生长种质保存的适宜方法提供理论依据。结果表明:生长延缓剂可提高试管苗SOD、POD活性,降低MDA含量,效应依处理浓度不同而异,分别在B9100~120 mg/L、CCC 50~75mg/L、PP3333~4 mg/L时达显著差异水平。恢复生长当代试管苗的SOD活性、POD活性以及MDA含量处理间表现相同。说明保护酶在一定程度上可减轻逆境对植物的伤害,延缓衰老。2品种相比,‘富士’试管苗SOD、POD活性及MDA含量变化对处理响应较快,且延缓保存效果优于‘乔纳金’。This paper explored the SOD and POD activity and MDA amount of 'Fuji' and 'Jonagold' apple explants in vitro during conservation period with B9,CCC,and PP333 supplied in subculture media,and at the stage after recovery to the normal growth status,in order to find optimal methods for germplasm preservation.The results showed that the SOD and POD activity increased and the MDA amount decreased with B9,CCC,and PP333 supplied in subculture media.The efficacy varied with the concentration supplied and the significant difference was found when the concentration reached 100-120 mg/L of B9,50-75 mg/L of CCC,or 3-4 mg/L of PP333.There was no significant difference among treatments in terms of the SOD and POD activity and MDA amount at the first subculture period after recovery from retardant growth preservation.It demonstrated that the protective enzymes could mitigate injury caused by stress to a certain extent and slow down aging process.The plantlets of 'Fuji' responded faster to the treatments in terms of the changing of SOD and POD activity and MDA amount,and showed better efficacy compared to that of 'Jonagold'.
