摘 要:目的:探讨小青龙汤对哮喘大鼠气道平滑肌细胞增殖的作用机制。方法:40只清洁级雄性SD大鼠随机分为正常对照组、哮喘模型组、地塞米松组和小青龙汤组,每组10只。观察哮喘大鼠气道平滑肌形态学变化;采用SYBR Green Real Time PCR测定哮喘大鼠肺组织中ERK1、ERK2和Cyclin D1的mRNA表达变化。结果:哮喘模型组肺组织中ERK1、ERK2和Cyclin D1的mRNA表达显著高于正常对照组(P<0.01),小青龙汤组和地塞米松组肺组织中ERK1、ERK2和Cyclin D1的mRNA表达显著低于哮喘模型组(P<0.01)。结论:小青龙汤抑制哮喘大鼠气道平滑肌细胞的增殖可能是通过抑制ERK1、ERK2的磷酸化和降低Cyclin D1的表达实现。Objective: To explore the mechanism of Xiaoqinglong decoction to intervene the proliferative effect of airway smooth muscle cells in the rat asthma model.Methods: Forty clean grade male SD rats were randomly divided into 4 groups,that was,the normal control group,the asthma group,the Xiaoqinglong Decoction group and the Dexamethasone group.Observing the change on morphologic of airway smooth muscle cell in the rat asthma model;The mRNA expression change of ERK1、ERK2 and Cyclin D1 in the lung tissue of all groups were detected by SYBR Green Real time PCR.Results: The mRNA expression of ERK1、ERK2 and Cyclin D1 in the lung tissue of asthma group was significantly higher than that of control group(P〈0.01),the mRNA expression of ERK1、ERK2 and Cyclin D1 in the lung tissue of Xiaoqinglong Decoction group and the Dexamethasone group were significantly lower than that of asthma group(P〈0.01).Conclusion: There is a possibility that Xiaoqinglong Decoction can inhibit the proliferation of airway and smooth muscle cell in the Rat Asthma Model,which can be achieved by inhibiting ERK1,ERK2 phosphorylation and reducing expression of Cyclin D1.
