鸡胚盲肠上皮细胞体外培养纯化方法的研究

摘　　要：为探讨鸡胚盲肠上皮细胞体外培养的纯化方法,采用低速离心去除单细胞、差速贴壁去除成纤维细胞纯化上皮细胞,并对培养细胞的纯度进行了测定。结果表明:采用1200r.min-1 5min的离心条件,利用贴壁差细胞种植1.5h后倒瓶的方法可去除部分单细胞和大部分纤维细胞,细胞种植第3天～第11天纯度可达72.00%以上。提示低速离心结合差速贴壁法可达到纯化盲肠上皮细胞的目的,可为试验研究提供较为理想的细胞模型。To investigate the methods of purifying the cecum epithelial cells cultured in vitro from chicken embryo,the cecum epithelial cells were purified by low-speed centrifugation to eliminate the single cells and adherent purification method to eliminate the fibroblasts.The epithelial cell purity was then measured.The results showed that some of single cells and most fibroblasts could be eliminated by centrifuging for 5 min with the speed of 1200 r·min-1 and changing container 1.5 h after cell planting basing on attachment gap.And the purity was above 72.00% from day 3 to 11 after planting.All the above experimental facts revealed that low-speed centrifugation combined with adherent purification could purify cecum epithelial cells,which provided a better ideal cell model for experimental research.
