摘  要:目的考察注射用美洛西林钠在4种临床常用输液中的稳定性。方法按临床常用剂量配制美洛西林钠的0.9%氯化钠注射液、10%葡萄糖注射液、5%葡萄糖注射液以及5%葡萄糖+0.9%氯化钠注射液,考察上述4种配伍液于0、0.5、1、2、3、4、5、6 h的pH值、颜色和澄明度外观变化。用高效液相色谱法考察在上述时间点下,配伍液中美洛西林钠的含量变化。采用BDS-C18(250 mm×4.6 mm,5μm)色谱柱,以磷酸盐缓冲液(pH值为5.8)∶甲醇(60∶40)为流动相,检测波长:210 nm,柱温:30℃,进样量:1μL。结果以本实验所建立的色谱条件对美洛西林钠进行检测,专属性好,能达到基线分离,美洛西林钠的线性范围为1~12 mg/mL(r=0.9999),日内精密度RSD<2.0%,4种输液配伍液在6 h内峰面积均无明显变化,变异系数RSD均<2.0%。结论美洛西林钠与0.9%氯化钠注射液、10%葡萄糖注射液、5%葡萄糖注射液以及5%葡萄糖+0.9%氯化钠注射液配伍后稳定,临床上可配伍使用。Objective To study the compatible stability of mezlocillin sodium for injection with four infusions.Methods According to the clinical common dose,the solution of mezlocillin sodium with 0.9% sodium chloride injection,5% dextrose injection,10% dextrose injection,5% dextrose +0.9% sodium chloride injection were prepared.The pH value,color,external appearance changes(color and clarity)and the content of mezlocillin sodium in the four compatibility solutions were determined at 0,0.5,1,2,3,4,5 and 6 h.The BDS-C18(250 mm×4.6 mm,5 μm)was used,phosphate buffer liquid(pH 5.8)and methanol(60∶ 40)was the mobile phase,and the detective wavelength was 210 nm with the flow rate of 1.2 mL/min,the column temperature was 30 ℃,and the inpouring volume was 1 μL.Results This method could achieve baseline separation.The linear range was within 1~12 mg/mL.The intra-day RSD was less than 2%.Peak area was relative steady within 6 h and RSD was less than 2%.Conclusion The compatibility of mezlocillin sodium with 0.9% sodium chloride injection,10% dextrose injection,5% dextrose injection and 5% dextrose +0.9% sodium chloride injection during 6 h is stable respectively,and it can be used for clinic.
