摘 要:目的探讨联合应用sIL-5Rα及sIL-13Rα2对哮喘小鼠IgE、INF-γ水平的影响。方法 50只BALB/c小鼠随机分为正常组、哮喘组、sIL-5Rα治疗组、sIL-13Rα2治疗组及联合sIL-5Rα、sIL-13Rα2治疗组(简称联合治疗组)。哮喘组及治疗组用鸡卵蛋白(OVA)和氢氧化铝致敏,用OVA激发,建立小鼠哮喘模型,正常组用生理盐水代替,其中sIL-5Rα治疗组、sIL-13Rα2治疗组及联合治疗组分别于激发前30min腹腔注射100μgsIL-5Rα、sIL-13Rα2及联合应用sIL-5Rα及sIL-13Rα2各100μg,正常组和哮喘组激发前30min腹腔注射相同体积生理盐水。比较各组支气管肺泡灌洗液(BALF)及血清IgE、INF-γ水平变化。结果与正常组比较,哮喘组BALF及血清中IgE含量明显升高,而INF-γ含量明显降低(P<0.01);联合治疗组可有效降低BALF及血清中IgE含量,同时可升高INF-γ的含量,与哮喘组比较有显著性差异(P<0.01);与单独治疗组相比,联合治疗组亦具有显著性差异(P<0.01)。结论联合应用sIL-5Rα及sIL-13Rα2治疗哮喘小鼠,可明显降低BALF及血清中IgE含量,同时升高INF-γ含量,从而达到缓解和治疗哮喘的目的。Objective To investigate the effects of sIL-SRcα and sIL-13Rα2 on IgE and INF-γ in asthmatic mouse. Methods A total of 50 BALB/c mice were randomly divided into 5 groups: the normal group, the asthmatic group, the sIL-5Rα treatment group, the sIL-13Rα2 treatment group, the combination of sIL-5Rα and sIL-13 Rα2 treatment group (or the combined treatment group). Mice in the latter 4 groups were sensitized with ovalbumin(OVA) and AI(OH)3, and challenged with OVA to establish asthmatic models, while mice in the normal group were treated with saline. Mice in the sIL-SRα treatment group, the sIL-13Rα2 treatment group and the combined treatment group were injected intraperitoneally with slL-5Rα( 100μg), slL-13Rα( 100μg) and the combination of sIL-5Rα ( 100 μg ) and sIL-13Rα2 ( 100 μg) 30 min before challenged, while mice in control group and asthmatic group received saline. Then the level of IgE and INF-γ in BALF and serum were measured by ELISA. Results Compared with the normal group, the level of IgE in the asthmatic group were higher, while INF-γ were lower ( P 〈 0.01 ) ; Compared with the asthmatic group, the combined treatment group can effectively reduced BALF and serum IgE level, and increased INF-γ level (P 〈 0.01) ; compared with single treatment, the combined treatment group also has significant differences( P 〈 0.01 ). Conclusion Combined with sIL-SRα and slL-13Rα2 to treat asthmatic mouse model, can significantly reduce IgE levels, and increase INF-γ levels, so as to achieve the purpose of mitigation and treatment of asthma.
