摘 要:目的:研究2型糖尿病患者牙槽骨成骨细胞生物学特异性。方法:采用组织块法进行体外培养2型糖尿病患者和正常人牙槽骨成骨细胞,倒置显微镜观察细胞的形态,用碱性磷酸酶(ALP)、茜素红(Alizarinred)染色法分别进行细胞成骨特性的检测,噻唑蓝(MTT)比色法检测细胞的增殖趋势,用酶动力学、放射免疫及酶联免疫分析(ELISA)方法分别进行碱性磷酸酶、骨钙素(BGP)和Ⅰ型胶原(COL-Ⅰ)浓度的检测,结果:在同等培养条件下,二者的形态无明显差异,但2型糖尿病患者成骨细胞较正常人成骨细胞生长慢、活性弱,矿化结节形成少;2型糖尿病成骨细胞上清液中ALP、BGP及COL-Ⅰ浓度均较正常人低(P<0.05)。结论:2型糖尿病患者牙槽骨成骨细胞增殖较缓慢,特征性分泌物含量较正常成骨细胞低。Objective: To study the biological feature of alveolar bone osteoblasts from type 2 diabetic patient. Methods: Osteoblasts from a type 2 diabetic patient and a healthy subject were respectively cultured in vitro with tissue explants adherent method. The morphology of the cells was observed under phase contrast microscope. MTT assay,ALP staining, Alizarin red staing, ALP, BGP and COL-Ⅰconcentration examinations, radio-immunoassay and ELISA method were performed to detect the osteogenic characteristics of the cells. Results: The cells of the 2 groups showed no significant difference in morphology. However, the alveolar bone osteoblasts from type 2 diabetic patient had slower proliferation, lower activity of calcium nodule formation, lower concentration of ALP, BGP and COL-1 in supernatant of cell culture than those from normal control(P0.05). Conclusion: The alveolar bone osteoblasts of type 2 diabetic patients can be cultured in vitro, but with slower proliferation and lower osteogenic activity.
