摘 要:[目的]观察健脾补肾方对腺嘌呤所诱发大鼠肾间质纤维化的治疗作用.[方法]灌胃给予200mg/(kg.d)腺嘌呤24d建立大鼠肾间质纤维化模型,将实验动物分为正常对照组、模型对照组、西药组、健脾补肾方小、大剂量组.建立模型后治疗第4周末检测大鼠血清尿素氮(BUN)、肌酐(Scr)、Ca和P含量.取肾组织行HE和直接红染色,观察肾组织的病理变化和纤维组织增生程度,采用免疫组织化学SP法观察肾组织中α-SMA和Ⅳ型胶原的表达.[结果]与西药组比较,健脾补肾方大剂量组大鼠血清BUN与Scr值明显降低,Ca与P平衡改善(P<0.01);西药组及健脾补肾方小、大剂量组肾组织病理改变及间质纤维化减轻,胶原纤维、α-SMA和Ⅳ型胶原阳性面密度明显低于模型对照组(P<0.01,P<0.001).[结论]健脾补肾方可改善肾功能,减轻腺嘌呤诱发大鼠肾间质纤维化,其机制可能与通过抑制肌成纤维细胞的增殖和活化,减少细胞外基质合成和分泌有关.OBJECTIVE To investigate the effects of Jianpi Bushen Fang (JPBSF) on adenine-induced renal interstitial fibrosis in rats. METHODS The model of renal interstitial fibrosis was induced by intragastric administration with adenine 200 mg/(kg · d) for 24 days. The rats were randomly divided into normal, model, western medicine, low and high dose of JPBSF groups. After treatment 4 weeks, the serum contents of BUN, Scr, Ca and P were detected. The pathological changes and proliferative degree of fibrotic tissue were observed by HE and Sirus red staining. The expression of α-SMA and type 1V collagen was determined by immunohistochemical SP methods. RESULTS The serum contents of BUN and Scr were decreased significantly, and of Ca and P were improved more well in the high dose of JPBSF group (P 〈0.01) than those in the model group. Pathological changes and RIF were alleviated significantly, and the areas of fibrous tissue, positive cells of α-SMA and type Ⅳ collagen were decreased markedly in the groups of western medicine, low and high dose of JPBSF (P〈0.01, P〈0. 001). CONCLUSION The JPBSF can improve the renal function and alleviate adenine-induced rat RIF, and its mechanism maybe relate to inhibit the activation and proliferation of myofibroblast.
