摘 要:将半胱氨酸-壳聚糖复合膜和金纳米粒子相结合,研制了一种禽流感病毒H9电化学免疫传感器。用循环伏安法对电极修饰过程中的电化学行为进行了表征,研究了纳米金的增效作用,考察了该传感器的峰电流与禽流感病毒H9浓度之间的关系。结果表明,纳米金能促进电极表面电子的传递,在最佳固定抗体浓度下,该传感器的氧化峰电流与H9抗原浓度呈线性关系,线性范围为1.0~170.0 ng/mL,检出限为0.4 ng/mL(S/N=3)。并且,应用该传感器对鸡的尿囊液实际样品进行了分析,其结果与血凝抑制试验基本一致。An amperometric immunosensor based on L-cysteine-chitosan(Cys-CS) and gold nanoparticles(AuNPs) has been proposed for the determination of avian influenza H9.The fabrication procedure of the immunosensor was characterized by cyclic voltammetry(CV).The synergy of gold nanoparticles on the immunosensor and the relationship between the current response of the proposed immunosensor and the H9 antigen concentration were discussed.Results showed that gold nanoparticles could promoted the electron transfer on the surface of gold electrode.Under optimal concentration of antibody,the oxidation peak current of this sensor was linearly correlated with the concentration of H9 antigen from 1.0 to 170.0 ng/mL,and the detection limit was 0.4 ng/mL(S/N=3).Moreover,real allantoic fluid samples of chicken were examined,the results were basically consistent with hemagglutination inhibition test.
