摘 要:目的研究pep-1-sod对缺血再灌注沙鼠脑组织凋亡相关蛋白表达的影响及其意义。方法将84只蒙古沙鼠随机分为假手术组、缺血再灌注组、pep-1-sod组及sod组,采用免疫组织化学方法测定各组不同时相bcl-2及caspase-3的表达。结果与假手术组及pep-1-sod组相比,缺血再灌注组及sod组在不同时点海马CAl区锥体细胞bcl-2阳性表达细胞数显著减少(P〈0.05或P〈0.01)。再灌注后6h,12h及24h,pep-1-sod组的caspase-3阳性表达率均显著低于缺血再灌注组及sod组(P〈0.05或P〈0.01)。pep-1-sod组与假手术组在不同时点的海马CAl区bcl-2及caspase--3的表达之间未发现显著差异(P〉0.05)。结论pep-1-sod可通过促进脑组织bcl—2蛋白表达,抑制caspase--3蛋白表达来实现抗凋亡作用,进而保护脑组织。Objective To study the effect and significance of pep--l--sod on apoptotic protein expression of brain tissue of Mongolian gerbil undergoing cerebral ischemia-- reperfusion. Methods 84 Mongolian gerbils were used to produce animal model and randomly divided into 4 groups, sham--operated group, cerebral ischemia--reperfusion group, pep--1- sod treated group and sod treated group. The positive expression rate of bcl--2 and caspase--3 in every group was determined by immunohistochemis- try during at 2,6,12,24,48 h after operation. Results The positive expression rate of bcl--2 in hippo- campus CA1 in cerebral ischemia--reperfusion group and sod treated group decreased significantly when compared with pep--1- sod treated group and sham--operated group during different phase (P 〈 0.05 and P 〈 0.01, respectively). The positive expression rate of caspase-- 3 in hippocampus CA1 in pep-- 1 --sod treated group decreased significantly when compared with cerebral ischemia--reperfusion group and sod treated group at 6,12,24 h after reperfusion (P 〈 0.05 and P 〈 0.01, respectively). No differ- entiation of positive expression of bcl--2 and caspase--3 in hippoeampus CA1 were found between pep-- 1--sod treated group and sham-- operated group (P 〉 0.05). Conclusions Pep-- 1 -- sod can cross through blood brain barrier and has anti--apoptotic effect on neuron undergoing ischemia--reperfusion. The potential mechanism is to promote the expression of bcl-- 2 protein and inhibit the expression of caspase--3 protein.
